Saving a contaminated jar?
    #1456983 - 04/14/03 12:04 PM (21 years, 8 days ago)

My dog has a jar of pes Amazonian that developed a yellow spot right next to a large mycelium patch.  he wants to save this jar because it is the only one in the batch that has shown growth so far and it might be the only one.  He injected an 6cc:1cc water and h202 mix onto the yellow patch.
---is it possible for the mycelium to simply overgrow the competitor and destroy it?
---Can my dog open the jar and take some mycelium from the other side of the jar, mix this up in a jar of water, wait for it to settle, then make a mycelium syringe which should indefinetly colonize multiple jars quickly?

My camera is out of bateries, but i may have a picture later today.  thanks in advance, can't wait to hear the replies

Yes, but it doesn't usually work out that way.]

You can, but chances are you will get even more contamination doing that.

The best thing you can do is just keep an eye on that yellow spot and hope for the best. Are you sure it isn't just mycelium piss?

What about transferring mycelial growth to peroxidated agar... wont save the jar but it will save the culture..

mycelium piss is yellow liquidy excreation, but this jar is only a week old, and the spot is round and probably growing...

why wouldn't mycelium from the other side of the jar be good for a mycelium syringe? what if i transfered the mycel. to a honeywater jar with some peroxide in there. Shouldn't the peroxide take care of the yellow mold if there even is any on that side of the jar?

was the injected peroxide in the jar a good idea? has anyone done that before?

So this yellow liquid excretion isn't harmful? It's just a byproduct of the mycelium?

No, it isn't harmful by itself but its acumulation leads to a proper environment for contamination, i.e. Anaerobic bacteria.

Yes, it is.

As others said, yes it's possible.

Well, it actually depends on the nature of the contamination and your skill handling this task but it's possible. Don't take the cake out of the jar, just cut a small healthy portion of the mycelium carefully and dunk it in a h2o2:h2o solution. Then the fastest and safest way would be using the mycelium directly has spawn in new prepared jars, you do this by using a glovebox, cutting the mycelium inside the glovebox and doing the transfer with open lids.
Read our faq on the issues glovebox and mycelium transfers.

Not a good idea.
You're messing with the water content which is no good. Also the contaminant gets exposed to the perioxide just in a short period of time, just enough to cause superficial damage, nothing more.

Thanks for the info dude. I think my dog wants to try dextrose water inoculation in a glove box perhaps. Do you know any good ratios of dextrose to water? hes gona put 1.5 tablespoons of "Equal" into a 1/2 pint jar of water then boil it for a half hour. he cant get any honey right now, but just plain dextrose should be fine right?

Sorry but i've never worked with dextrose before.
Why don't you just make a direct transfer ? Then after growing, make prints right away. I don't see why you want to have all that work, plus, dextrose as honey water is very prone to contams. Your call.
