spores from dried caps

I spent some time looking and looking but had no luck finding a certain carmel cap that I have become fond of. Ps. baeo.
After thinking things through I decided to try using some dried caps since that is all I had on hand.
I put a pint canning jar filled with water in the mocrowave and let it boil for about ten minutes. Then , I poured off the water until there was about 1/2 inch in the bottom of the jar and placed the lid back on while it cooled.
I put three caps into the jar after things were cool and let that sit a few hours.
Next, I made sure the lid was tight and shook the jar with all my might a couple times.
Holding up to a lamp you could see the nice brownish purple coloration of the water.
Now I know things could be contaminated but getting the spores was the issue and a couple transfers is no problem.
So, thats all except drawing up some of the dark liquid .

are you going to inject these in jard directly or transfer to agar ?

Well people , the test did'nt go too well. I erred. Seems there was too much bacteria on the dried cap and that just exploded with growth.
I think I will just crumple up a dried cap next time and go from there. I know that will work because I've done that before.
I guess I just love to experiment.

you would need to use the agar thing and isolate mycelium to another plate,, and transfer afew times to eliminate the contams !!!! this WOULD surely work.!

Crumbling the cap .. well the contams get crumbled with it!! no use!

you could try rinsing with some h202, it wouldn't hurt the spores much

I was thinking rather than initiating on agar,how about honey water? It is somewhat antibiotic and may inhibit a portion of the bacterial load.Once an obvious mycelial mat forms pull a portion and plate and isolate from there.I do not believe one will entirely eliminate an agar phase unless a selective toxin is used for bacteria that will not kill the spores.I do not know of such a substance,perhaps one of the premeds or biologist about may know of something.So what do the experts think? Would a weak honey sol be more inhibitory to certain bacteria and be of benefit or would it just proliferate the contams worse? How about sandwiching the culture under peroxidated agar after isolating it from the honey water? Again many questions for a hobby :grin:WR

ok then

why even bother with honey water, if you are going to go agar anyways???

Total waste of time man!

I would think that if honey benefits to mycelium, it will to contams as well!!

Key:
Honey is a somewhat selective media and may inhibit a portion of the bacterial load, therefore plating a pure culture in fewer isolations theoreticly.

In actual fact any water medium aids contaminants to no end as it lets any latent spore spread throughout the medium. agar is much better in inhibiting or isolating nasties, but ofcourse as the Austrian said H202 will kill spores.
