getting spores from dried specimen?

is there a way to get spores from a dried specimen? Cause, i could just buy an 1/8th and get the spores out of them...

I remember reading somewhere that you could do it, by putting a drop of water in a bowl, and put it over the mushrooms, rehydrating it, but i can not find any solid information to back it up?

hmmm.. you probably could get the spore, and even germinate it. But I dont know how sterile that will be. you'll probably grow more contaminations then anything else. Anyone else have a thought on this?

he could:
a) inject some sterile water on the gills, and suck it back in, hoping for no contaminants -->chances:poor
b) extract a gill and put it on antibiotic agar -->chances: fair

I have done it several times by extracting a bit of gill tissue and placing it on antibiotic agar. Expect contamination but with a little work you can clean up the culture.

Sorry for stealing the thread but:
What kind of antibiotic is used in antibiotic agar?

I had to eat some antibiotics, could these be used as i have one left. On the package it says antibiotic - cefalexin. monohydr. 750mg.
Any ideas if these could be used? Otherwise i guess i have to try!
Could be a longshot though... :tongue:

thats what my jars are in the middle of right now. ive only had contaminants in one, but they are doing shitty... and i hate them... wish they would colonize. i would tell you to expect slower growths. if your going to do it let it hydrate for at least like a few hours it makes it easier to suck up the longer you let it soak, and allows what i would think the spores to be released from the gills. they are pretty chunky pieces but soak them you can suck them up. it worked for me, its just going slow. but maybe if you stick it in a honey water solution, it could culture and you could suck it up and have lots of mycelium already going and not have to worry, you could judge contaminants etc. i dontk now if the honeywater thing works, im trying it today but in theory i would expect it to work, you could also try agar. check the tek

Which is only effective against bacteria which are not usually as much a problem as molds

gentamycin sulfate is used because it can handle being pressure cooked without breaking down, I don't know of any other antibiotics which can claim this.

Yes that is pretty amazing, the ones i was referring to say to store in a temperature under 77F, so i guess PC:ing that stuff is out of the question  :smirk:
Although my first thought was to add it after PC:ing (as with peroxide).

Molds can be controlled with very small amount of benlate (benomyl) in the agar 1/100 gram per liter. Higher amounts slow the mushroom mycelium so careful measuring is required.

For serious contamination a combination of gentamycin and benlate will inhibit molds and bacteria.
