I still don't see how that's better than

1 to 160+1
161 to 25921+161

I mean, if we're gonna talk theoretical yields here myc water outpaces g2g at an astronomical rate, even if you can get in 3 g2g cycles per each myc water cycle. Wouldn't senescence set in sooner in g2g as well?

I'm not trying to criticise here, I"m just trying to understand. If you're arguing from pure speed, g2g seems to win hands down, but for bulk myc water seems to be the clear winner to me.

You can do three G2G transfers no problem,  that is a 1000 jars from one.  It is faster and less chance of contams.  I don't know anyone who needs more than 1000 jars of cubes.  In the long run G2G is the wiser choice unless you can afford to lose jars to contams and senescence should not be in this because it is the same myc if any thing the myc water would break the myc down faster than g2g.

But if you want bulk and speed, instead of starting with one jar, you can start with 100. That way you'll have 10000 in less than a month...

Is it enough or do you want some more spawn?...

well in my state if you get caught with more than 400 gram you goto prison for no less than 5 years up to 99 years. So i think 1000 jars would be enough for safety reasons...

This is not technically correct, it's 1000 jars from 101 jars. I don't see how g2g has less chance of contams when you're opening the jars to the elements whereas myc water is all done from within enclosed sterile environments. From that it would seem to me that the opposite would be true.

I agree that the numbers are ridiculous on both counts.

I suppose the senescence argument hinges upon whether senescence depends on the amount of expansion the mycelium does or the actual age (in days) of the mycelium.

Even just using a simple GB I have never had 1 jar lost to contam when doing g2g it is just to fast for contams to set in.
While using myc water I have lost several,  the extra moister and slow recovery time just doesn't work for me.  I am not hooked on the GLC,  yes it's easy for a noob but not worth the effort.IME
So I am done with this thread if you like GLC use it but if you have a glove box try g2g.

No worries mate, I'm just trying to educate myself as well as I can about these things and the voice of experience is always appreciated. As has been proven many times, what seems to make sense in this hobby is not always what is right, and the more info a person can get the better. Thanks.

i think senescence is more to do with expansion than age.

Cell lines can only divide so many times before things slow down, and go wrong.

The main advantage of G2G is colonization speed. And that may be the only advantage...

With GLC the mycelium is torn up and shred apart really bad, to the point that it's small enough to be sucked up into a needle. After this much abuse it's going to take the mycelium a couple days to recover and start growing again.

With G2G the mycelium isn't damaged nearly as much as it is with GLC. So it takes less time to recover, and starts growing faster, resulting in faster colonization time.

I've tried 2 LC's and I've never had a successful grow from one.

I've wasted a ton of resources from a LC of PE I was trying to grow out.

I sectored out my cultures drop the agar piece in the grains and g2g that. By doing this I am 100% confident my culture is clean and I've never had a contaminant in my g2g's from doing it that way.

That's getting it done with confidence

I've done both and I would prefer to do a G2G over the grain LC.

I use regular LCs more often than both because it's simple, effective, and I don't need a GB at all. Just open air work. Ya gotta be clean with the syringe and all that though.

Why would you need a glovebox for myc water (grain lc) and not normal lc?

You're right. You don't need a GB for grain LC.

I just meant that I prefer to use regular LCs over grain LCs because it's easy, doesn't need a GB, and LCs are self regenerating. Grain LCs are more like a MYC extraction so to speak. You rob the MYC from a jar and that's that ... well at least that's how it worked for me. I've tried letting the donor jar recolonize, but it just didn't happen for me. I quickly switched back to regular LCs.

Grain LCs are worth trying. Everyone has their own preferences and that might be the way to go for some.

Wow ... I've never tried agar, but have done real well with LCs. Maybe it was just the LC recipe you used. I like dry malt extract with karo, it's the best recipe I've ever used. It's like liquid gold to me now  I recently cloned a fruit (using a GB) and dropped 4 pieces of tissue into 4 pints of malt/karo LC and 100% success ... no contam at all.

I've inoculated LCs using PE spores and that shit sprouted overnight literally!

Here's a pic just after inoculation with PE spores. The flecks are just malt sediment:

This is a pic taken just 2 days later:

5 days later:

Here's the recipe if you're interested. If you can do agar then I know this will work for ya.
500 ml pure water (I used RO/DI water)
10 ml Light Karo
1 g Extra Light Malt Extract

Here’s a breakdown of the same recipe for different volumes

600 ml pure water
12 ml of Light Karo
1.2 g Extra Light Dry Malt Extract

250 ml pure water
5 ml of Light Karo
0.5 g Extra Light Dry Malt Extract

I pressure cooked the jars for 30 min @ 15psi

I purchased 1 pound of the extra light dry malt extract on eBay for about $12.00 including shipping. I paid too much, but it was well worth it because 1 pound will likely last a lifetime.

If you do the mycelium water tek, things might get messy. Definitely follow Agars tek to the T. I tried to do it on a larger scale, (fully colonised quart jars 3/4 full), and it was hard as shit to get the liquid back out withou making a mess. The liquid I got out didn't work very well either.

If I were to try it again I would use pint jars a little less than half full of colonised grain. That way theres plenty of room to shake the mycelium off the grains. Getting the water back out can be tricky though; for me at least since I dont have all the materials to make a good lid. Or maybe I'm too lazy to do so...

Anyways, I'm all about the G2G. It works great for me. It takes off fast and it's easy as hell to do. There is a chance for contamination but I havent had a bad jar in quite some time now, and I dont follow the strictest technique. I use my oven door with the heat on low and lots of oust in the room.

G2G is the shiznit. But so is a good LC to begin with.

This picture is of a 4 day old jar knocked up with 7cc's of an old ass LC I've been keeping at room temperature for 4 months now. Works very well, espicially considering I thought it wouldn't do shit since it hasnt been refrigerated.

Just my 2 cents

That is an unbelievable amount of growth for just 4 days ... WOW! It usually take me a day or two to see growth on my grain from the LC inoculation.

I like that so many people are pointing out that they've had a good success rate with g2g.  People seem afraid of it; they need to open a  sterile jar.  But glovebox work is really not too difficult.  G2G is awesome, you can inoculate however many jars, 7 quarts is one batch in my case, so I choose the jar that colonizes the quickest for the grain transfer.  The "quickest" colonizer isn't necessarily the best fruiter or most potent, but it's a good route if you're just starting.  I prefer seeing normal MS projects, but I've gone that route before with a successful project. ~Pixie~

It took 2 days to show. But since then... BOOYOW! Mind you thats a pint jar with 7 cc's of LC. I shook it too. I don't understand why some people use 1 or 2 cc's for inoculation when you have a couple hundred at your disposal. I personally like things to move quick. I hate waiting weeks for jars to colonise. FUCK THAT!

And I'm gonna take that pint jar and do a G2G to my quarts. I'm going to knock some up with the LC as well. Both at the same time so I can see if one way is faster than the other. I doubt there will be much of a difference in colonisation times. I think it has more to do with the amount of inoculate or colonised grain you use from the get go.
