experiments results look good. here is what i did.

MEA was sterilized solely in microwave (3ms high + 13ms defrost) on sept24;
H2O2  was added at standard concentrations;
spores (from spore print) were dropped on sept25 on wetted piece of cardboard which acts as C2O2 barrier for germination;
the petri was cheked today, oct21, showing aggressive healthy rizhomorphic growth

alltogether, it seems this tek works well, and i recommend it to all those like me working without a HEPA :smile:

did you use more than one disc per plate? the plate on the right looks like it has several germination points. or maybe some spores somehow survived the h2o2?

great pics. i love the microwave for doing agar. i have some millet just innoced that i fractionally sterilised in the microwave.

blackout, could you elaborate further on that. iyou mean you sterilize the grains in the microwave? how do you do that? i am a big microwave fan, and would like to put together a complete, solid "microwave tek": perhaps we could join forces! tell me everything you do with the microwave (in a mushroom growing context, that is) :laugh:

Champion des Champignos (or however you spell it) has also done this sucessfully. I think also zeronio. Help out here guys!

yes, i found the type of cbd is important. with the champ's help, i discovered the best is non-corrugated, the type on the back of a regular notes pad. also, the smaller the cbd inoculation point, the faster the myc will spread to the c2o2 agar and take off beautifully

i boiled millet in a 2 jars with a filter lids for 20mins. let it cool and drained off the excess water and rinsed them and drained again. i didnt drain them for long since i knew they would lose more water on subsequent boilings. i put them both in for 8 mins together on full. when i take them out i shake them and wrap them in thick towels which keeps them warm for the rest of the day. i dont know if i really need to do this but the faq talks about having them at 100C for 30mins. i have tried and failed before, i think i had a contamed syringe, another time i had dodgy grain which mould even had a hard time growing on. i hope this millet is ok. another idea i had was soaking rye/millet for 2 days in initially boiling water then loading it in a jar and incubating for a few days. by then all the endospores should be germinated (maybe) then i was considering microwaving them until they were bone dry, maybe an hour or so. then i would rehydrate them with liquid innoculant. this would take a lot of experimenting but would be easy if done with small jars. you would need a scale to weigh grain to see if it has totally dried. i.e say you normally add 100ml water to 100g rye. and your jar is 200g. add your soaked rye in the jar and it should all weigh 400g nuke it until it weighs say 280g (moisture is in rye grain to begin with which anno has done measurments on). then innoc with 120ml of innoculant.
i also want to experiment microwaving substrates mixed with graphite which gets extremely hot in a microwave (hot enough to melt metal)

hi blackout, thanks for the info. it's all very useful, but i think we've got more work ahead to develop & refine a complete, succesfully tested, "microwave tek". let's keep moving on this. all experiences, ideas and suggestions on this are welcome

the problem with a deifinitive microwave tek is that every microwave is different. even ones with the same power rating will give different results. even different shaped jars with the same substrate in the same microwave will give differences. the differences are the final moisture levels. that is why i want to try out the method of totally drying out a substrate and the a subsequent exact rehydration with sterile water. the only problem i can see with this is a possible loss of nutrients.

blackout, please keep posted, i'm very interested in your experiments. does'nt soud to me lack of nutrients would be a problem, more difficoult how to rehydrate at the proper moisture levels

rehydrating is easy enough. just inject sterile water back into the jar and shake. if you have weighed the grain and are sure it is totally dried out then you can work out how much water you have to add. if this water you are adding is innoculant then you will see a very fast growth occuring. i do not know if the high heating will cause nutrients to degrade though..
