Azurescens on peroxidated agar?

Hi, I have just whipped up my first batch of 8 petris with: 0.5g nutritional yeast 4.5g agar agar 5.0g liquid malt extract 250 ml water (originally however I think a lot of it evaporated) After pressure cooking for 25 mins I waited til it reached 55 deg C, then added 2ml of 6% H2O2. I poured this into 8 90mm petris. Later I will innoculate each with a chunk of wood colonised with Ps. Azurescens. I just want to check that Azurescens does well on peroxidated agar, that I haven't used too much peroxide, and could someone please tell me the optimum incubation temperature for azurescens. Thanks a lot

P. azurescens does well on peroxided agar, yes, BUT:

this would be 8 ml of 6% peroxide for 1 liter or 16 ml of 3% per liter.
Wayne recommends 6-10 ml of the 3% per liter....

oops, anno is more precise, as usual. you might still be ok, although the C2O2 concentration is on the high side. are you sure it was 6% and not 3%, which is the standard at least here in the us? even at 3%, 2mls per 250ml of water is high

Yep, it was 6%. I will just mix up another batch in case the first doesn't do well. I have plenty of petris now.. the bastards wouldnt sell me less than 500! Thanks for the advice

Righto, thanks for the advice people. I have made another set of 8 with 1ml of 6% H2O2 (in 250 ml of agar). The plates are incubating at about 20-21 degrees C, is this okay for azurescens? How fast does azurescens mycelium typically grow?

Ive always used 3-4mls per 250mls of mea mixture and all my mushrooms have thrived. The mixture i use was recommended to me by a local spawn company and they have never had mutations as a result of this ratio. If your attempting to use virgin spawn or colonised woodchips or clones on your agar, i always up the h202 to about 4-5mls per 250mls. I never ever use less than 3mls of h202 for 250mls.

Hi, The azure is coming along well thanks. I'm incubating at about 22 degrees (I think), in my hot water cupboard. There is little difference between the correctly-peroxidated and over-peroxidated petris.
Most are fully colonised like this. I have sectored a few chunks out onto new petris to try and get some more rhizomorphic growth.
I have also inoculated a few jars of rye grain with agar wedges.
I'm not planning to fruit off the grain (obviously). I think I will spread it out in a tray, cover it with a layer of wet cardboard, wait til the cardboard is fully colonised then cover with soaked woodchips. Finding the right woodchips is a bit of a problem for me at the moment though. Can anybody tell me whether I should shake these jars or is azure mycelium too fragile? Thanks
