yes you can do a lc with the isolate. you want about six or so isolates to test. this way you can pick the best ones that fruit better with good pinsets ect. what ever traits you are looking for. look for AGAR AGAR in oriental\asian supermarkets and stores.

Once you have an isolated strain and you grow this fruit out if you do a spore print and then grow from these spores is this basically taking you back to square one or will the selective growth work done in the isolation carry over to the prints spores? Still trying to get my mind around it all.

That was what I though but wanted to check. what you are really working on in agar is mycelium growth traits is that right?

Can karo be substituted for agar if I can't find it, or is it only compatible with LC?

No u need agar you can order some online at Mycosupply one of our sponsors.. They carry it and it's not to bad of a price.. But you should be able to find agar agar at an asian market near your home..

This is why once you isolate your strain on agar you use it to make a master slant. This master slant can be kept in the fridge for a relatively long time.

Each time you want to grow you take a tiny piece of the master slant, grow it out on agar and then use that growth to inoculate say 3 or 4 grain jars. Then you do a grain to grain tranfer and turn those jars into many more (even up to 30/40) and use those at spawn.

So that way you can do hundreds if not thousands of jars from that single master slant before it is time to isolate another.

Karo is a nutrient, agar is a support media. It does the job water does in an LC. In fact you need to add food to your agar (Potato-dextrose, brewers malt, or others) or buy one with the food ready added.

A search using the keyword agar will give you a load of information on where you can get either lab grade or food grade agar and make the mix for your petri dishes.
