What is an isolated strain?

I'm currently on my first grow and have now come to the realization how unreliable and unpredictable multi spore is. I have fruits that could be skinny and tall and very short and fat growing off the same cake. From my understanding strain isolation is used to pick the best genetics but I truely don't know so fill in if you want. If it is in fact what I think how would I go about doing this? Can I use karo in place of agar?

You would need to start the grow on a petri dish, and transfer sectors to other petri dishes and as they show individual growth, transfer them again and again until you're down to single sector isolates.

You probably want to do 5 or 10 of them to that point, then grow each one and see how well they grow and how potent they are then keep the good one(s).

The other, and easier method is to grow a multispore batch then CLONE some fruits that have features you want.  First to sprout, biggest, shortest, tallest, thickest, etc.

This is different from isolating because each mushroom can (and probably does) have several sectors in it.

Ok, yeah thanks texts on isolated strains were the only ones I've never understood. I guess cloning would be my best choice for now. I'm really dissapointed in multispore and needed to move to something else.

If you want a true isolate gorw it out on agar.  If you want something better than ms, but are afraid to work with agar, do a LC clone.  It won't be a true isolate but it will exhibit the qualities of the cloned fruit.

I haven't done any agar work yet but I've been researching it and it doesn't look like it would be that much harder than doing an LC.  Plus, an advantage over LC is that with agar you can see a contamination on the plate, and transfer healthy myc to another dish, with LC you can't always tell if your LC is contaminated until you try it out.

Strain has two meanings.

You have your commercial strain which is B+, Golden Teacher, Penis envy etc

Then you have real strains. Real strains are a set of individual genetics that have been isolated by putting spores on agar, letting them grow, and taking little pieces of them and tranferring onto a new dish of agar and so on until no sectors appear. You will then have a puere strain, but you have to grow it out to test it to see if it has the characteristics you want, as you have no idea how that strain peforms.

This is a lengthy process.

A good alternative is to clone a fruit that you have selected for good size or prolific fruiting, or whatever you prefer. This does not give you a pure strain but it is prettty close, and it saves you the hasstle of testing ten isolates to find a good one.

don't mean to hijack but what it the real purpose of isolating from a clone. is it because each individual mushroom has separate sub strains in it? would different sectors from the same clone produce much different mushrooms ?

You could find different results yes.

Also, the mycelium from the clone will already be quite a few weeks old which means that you will be able to do less transfers from it than you would be able to do if you have a much younger master isolated on agar.

Does anyone have pictures of what sectors look like ?? I am getting ready to do some agar for the first time today after the purchase of a new tub for a glovebox.. But I am not sure what these sectors look like..

its possible to grow out a clone batch that has different qualities because of the other "substrains" (as i call them), if a different substrain becomes dominate in the clone grow. sorta like re-rolling the dice with less strains. but i guess its kinda rare that cloned batches produce different dominate phenotypes (traits). but not imposable.

I'm a cheap bastard and use a lot less. That stuff is so expensive.

I figure it costs about 5 cents to wrap a petri dish with parafilm.  That's pretty cheap insurance, imo.

The easiest way of getting better fruits (but also the most lengthy and inefficient.) is to grow from multispore, puck the biggest and fastest growing mushroom, and take a spore print. Then grow that print and choose the best one from that, and make another print, and continue the process for quite a few grows, within a few years of growing, you will have some very good mushrooms, but it takes much longer. If every time you just choose the best mushroom, eventually it will be better than a isolate, as you will be evolving the specie, not just choosing the best one and cloning it.

you can improve the genetics over generations but this by no means takes away the variation of multispore! you will still see poor pinsets and different looking and sizes of fruits. and potency with actives will still be a "roll of the dice"

Not exactly true.

I won't go into an explanation of fungal reproduction as it is a lot more complex than that of humans or plants and changes from genus to genus. You would require a solid background in cell biology to understand the process fully.

To give you a general idea though:

Fungi, when reproducing sexually (i.e. producing spores), undergo mitosis. Mitosis entails random gene crossover which means no two spores are identical and they contain genetic information from the different strains that made up that particular fruit.

Each time you then grow from multispore you are producing mycelium that contains the genetic makeup of all the different spores that produced hyphae and merged.

In essence, what this means is that even though by selective breeding you might be able to reduce the genepool somewhat, it will be practically impossible to eliminate all genetic variability.

Even spores from an isolate made on agar will have different genetics, admittedly a lot less variated than that of spores from a wild fungi.

Ok, so when you get an isolate with no sectoring would you do an LC to grow that mycelium out? Also, where can I get agar? This thread has really helped me out in understanding this topic. Thanks Guys!

You could make an LC or add an agar wedge to a jar of grain and then do grain to gran transfers to multiply that jar.
