Grade of Agar
    #873460 - 09/09/02 11:02 PM (21 years, 5 months ago)

I`ve done a search on the best possible grade of agar to use but all I can find is that it`s not necessarily important having a higher grade.
Does anyone know what the best possible grade would be to use for growing isolates.
I`m getting the agar from a lab so no-one give me an answer of "use the highest grade you can get" because well that could pretty much be anything.
Also does the higher grade agar not have any down sides??

If the answer is , use the highest grade, well then fine but if anyone has a possible grade table for me or knows of the grade which would be best, please reply.

the agar I use is from a health food store, so I suppose it's "food grade", and it works fine. As agar is essentially non-nutritive I don't see how super-high-quality grade agar could make much of, if any, difference.
Maybe it would have a higher tensile strength or something, but I don't think that's really anything to worry about in our line of work.

Microbiological grade agar.

" Most high-grade agar applications for microbiology and agarose require agars derived from Gelidium. Gelidium agars from different species and locales differ in their properties and not all Gelidium agars meet the specifications for microbiological or agarose applications. Lower grades of Gelidium agars are utilized in food applications. These microbiological and agarose agars are mostly Gelidium agars. Uses include: the preparation of culture media for bacteria, yeasts, and molds; plant and animal tissue culture; biotechnology; genetic engineering; DNA and protein separations; cloning of organisms; and the breeding of insects used in biological control. Other applications include its use in producing exact molds for manufacture of dental prostheses, reproducing sculptures and archeological pieces, reproducing fingerprints in police investigations and gelled electrolytes in the manufacture of leak-proof batteries. An estimated 100 mt, from the 10% noted above, is used in the production of agarose, a high-value, electrically neutral component of agar obtained through further refinement, which forms strong gels at low concentrations. Due to the special properties of agarose, it is utilized in DNA testing and provides an excellent medium for plant and animal cell cultures."

Thanks

My understanding is that as long as it is good agar it will work?
Because well as Champion said, most people use food grade agar.
Also Anno I see the site you posted refers to using bacteriological agar of atleast 800 gms/cm2.
So atleast I have that to work from.
I`m sure the lab will be able to use this info better.

If you are getting your agar from any type of normal lab it should be bacteriological/microbiological grade. At least in the labs I have worked in we have not used any lower grade agar. Sure you can get away with using food grade, but the other grades will give you firmer surfaces and less visual impurities, thereby making your analysis of your culture relative to contaminants easier.
