yes you can shake the jar, in fact do so before you make your syringe so that the mycelium is spread out through the solution. i use a leftover canning jar to do the same but with only one hole poked in the top which i uncover and place the syringe in to suck up the fungi. be careful if you take the whole top off, you could introduce contams.

This is what I usually do. I PC the liquid substrate in a 1/8 pint jar with a hole punched in the top (filled with polyfill.) What I do different is I cover the top with the rubber seal facing down (making contact with the glass lip.) This effectively seals the edge of the jar. There would really be no need to turn the jar over all the way even with a small needle. Just tilt it far enough so that you can reach the solution. If you try this without sealing the lip, it can leak on you.

I guess a bead of silicon might work. Try it and fill us in. My primary concern would be plugging the end of the syringe with a "core" sample.

They are all medical grade needles... unless your using a turkey baster You should be using a 16 or 14 gauge needle for your liquid culture, and it IS large enough to plug with the silicone.
I remove jar tops and keep a sterile (as can be) environment while I'm working. Havent had a problem yet. If your worried, you might consider using the oven tek to fill your liquid culture syringes.

The tip of a diabetic needle is very small in diam. the mcellium will get stuck in it, never mind the silicone. You need a larger gauge tip... 16 or 14 gauge. Check out a nearby farm supply store for them.
