Liquid Cultures (steps A - Z)

I've been reading alot on liquid cultures, but theirs are just too many scatterd ideas. Could somebody plz just clear some of it up.

I would like to know what to use as a liquid and where to get it.

How much to use.

Last but not least simple working technique.

If you could help me I would love you forever.

A *beginner friendly* method for making Liquid Culture

A simple LC tek by Otto with pictures (no PC required).

I have this crazy idea...

Can you make an LC and before it does what it does, could you pull it into the syringe and have it grow inside that???

I second this. Used this and variations of this since and it's made liquid cultures easy and painless.

I even microwaved karo/water for ~4 mins in a plastic container w/ a small hole poked in the lid and saran wrap around it, refrigerated it until it was cool, then used my LC I made w/ Otto's instructions to make a bunch more syringes easy. No contams yet w/ these.

I am very new and don't really know, but I wonder... what would be the point of doing this????

THe point is its much much much faster. Plus it gives you a chance to get more involved. I dont know about you guys but to me this is more fun than mario when it first came out.

OOPS, I didnt see that you quoted about the LC syringe.... I just want to know if its posible, that way if it does work, then you can grow them in the syringe and they are ready at any time that you need them instead of setting aside time to prepare them later.

its possible.  there's not really any gas exchange in that arrangement and it goes very slowwwwwly

another neat trick is to take an "empty" spore syringe, and suck up some sterile LC solution.  there's always enough spores stuck to the sides and plunger that it will eventually recolonize a nice little mini-LC :wink:  but again, it will take its good ol' time.  better to use a more traditional container w/filtered gas exchange

my favorite recipe is 96 grams/ML of water to 4 grams/ ML of honey. mix well when warm and pressurecook for 20 minutes and no longer otherwise the honey will carmelize.

there is no gas exchange in a syringe so no it can't grow. you can store a live culture in a syringe but not grow in one.

How do you know when there is contamination? Isn't it hard to spot? What do you look for?

you have to test the culture. the only contam i have ever spotted was black mold but trichoderma (green mold) is harder to spot.

Technically you don't need any gas exchange for a lc to grow.... but if you do this, and a big huge mass grows inside the syringe... you will not be able to chop the mycelium into bits so that it will be able to pass through the needle hole.

besides i want my lc to look like little snowflakes when knock'n up my jars. :wink:

my only advice...

KEEP IT SIMPLE.

You do that, and you'll have good luck with making and using LC's. The beginner tek is a good example of keeping things simple.

Use this tek and use honey and not karo,add 2 sharp shards of broken glass into each bottle to rip it up small enough to suck into a needle

I just drill a large hole in my quart jar and use a tyvek filter over it. You don't need to shake them, a quick swirl once a day is plenty.

Bah on Karo and Honey, both have failed me.
dry extra/light malt extract, and dextrose (corn sugar)1 tablespoon each into 32oz boiling distilled water, stir it up good, run it through a double or triple coffee filter, split into 2 quart jars, lid with an inoculation spot and a gas exchange spot, or use tyvek/micropore tape and only use 1 hole. Cover lids in foil, PC the jars for 20 mins at 15psi, let it cool naturally, wait at least 15 mins after the pressure drops to 0, and remove the jars, and foil.
Put a rubber banded coffee filter over the top of the jar as a dust cover if you like, but it does nothing for contam spores.

Don't forget the stirbar, or glass shards to break the mycelium up.
