Is it contaminated ?

Okay tonight I did one of the dumbest things in my life. I got my spore syringe out ready to innoculate. I pulled the tape off the cap and instead of just pulling it off, I unscrewed it !!!!!! I just wasn't thinking. The cap and hypodermic needle were seperated from the syringe. I realized immediately how stupid I was and screwed it back on. Did I for certain contaminate the liquid inside ? It was exposed to the air for maybe 3 seconds. Although my A/C and all the fans were off. I'm so bummed. I dipped the whole end into a glass of isopropyl alcohol and let it dry before injecting the jars. I have such a bad feeling that every single jar will be contaminated. What do you think ?

It's all academic at this point, because it either was or wasn't, but here are some things to consider: To put the needle on the syringe, initially, one must uncover both the end of the needle and the end of the syringe. (Unless, you happen to buy syringes that come with the needle already attached. I've seen those.) So long as you didn't touch the ends of the needle and syringe with anything, you maintained sterile technique outside of the fact that you opened the end. Exposing an area as small as that to the air for 3 seconds is giving bacteria very little chance to infect anything. Realizing that bacterial contamination rates will vary from building to building, and with different times of year, here's a test that we ran at our molecular biology lab: (10) 120mm agar plates were laid out without their tops for various times 15 seconds, 30s, 1m, 2, 4, 6, 8,10, 12, 15m. After looking at the colonies that grew on the plates, the average inoculation time for the lab was about 1 colony per 115cm^2 per 35 seconds. Quarterly, this average varies about +/- 5 seconds, but it's still a good estimate for where I work. Your miliage will vary, but I wouldn't be worried about contamination in a span of 3s/1cm^2 as compared to 30s/115cm^2, so long as you maintained an aseptic technique otherwise. TiFox

Why is that? It's a valid question, it's in the proper forum (although it could have gone either here or in contams as he was in the process of growing), and we have all done things without really thinking while we were doing them.

One last thing to note is this: If the luer lock end of your needle or the end of your syringe *did* happen to get contaminated with an airborne particle, it had only one place to go: out, most likely in your first bag. You will find out in about a week, though. I'd have smiles instead of worry. TiFox

Thanks for the words of encouragement everyone. I guess there is nothing to do but wait. I should see mycelium growth within a week ? If there is no funky stuff I should be out of the woods ?
