Needle Sterilization Question

Can I just burn rubbing or isopropbyl rubbing alcohol to get a sterile flame capable of cleaning the surringe tip?

Yup, that should work just fine, as long as the alcohol burns. A lot of the time rubbing alcohol is diluted with too much water for it to burn well. As far as I know, any alcohol should burn with a clean (aka non-carbon depositing) flame.

Isopropyl alcohol doesn?t burn clean.
Use ethanol if possible.

I just wipe mine with a paper towel soaked in 99% iso. alc. never had a problem

I do exactely the same. I bought an alcohol lamp, and one month later it is still sitting on my computer desk. Empty. Just wipe the injection site and the needle and then it is smooth sailing.

Wal mart has the 99% for a buck or two-it's in the pharmacy area. Just put a couple of drops on the bottom of an upside downshot glass and fire it up.
Wipe needle with paper towel soaked in 99% before innoculation.
You can also swab the innoculation site if you want to be extra safe.

my friend just uses a bic lighter then wipes the soot off with rubbing alcohol. just takes a little longer to get the needle red hot.

I don't know why you people do this to your needles. They are sterile when they come out of the package. If you just stick it on yuor syringe and move quickly from jar to jar innoculating, you dont' have to clean it.

Surely your not alchohol wiping and flame sterilizing between each innoculation, thats like 4X per jar times 10 jars= about an hour to innoculate a few jars? lol!

If you think your needle is contaminated each time you stick it into the substrate, then you must be doing something wrong. That is supposed to be a sterile substrate, you're not contaminating your needle by sticking it in and it doesn't need to be cleaned each time.

Of course, if you are just being thrifty and reusing old needles, then fine but really, they only cost a few cents. Just use a new one.

it doesn't make sense to sterilize after each hole, but sterilizing between jars is just being extra safe.

people can take all kinds of non-sterile short-cuts and many will do fine. but some will not. the extra precaution increases the chance of success with very little inconvenience.

a person working alone may have to put the needle down while de-taping or re-taping holes. who knows what airborne contams may be falling on the needle when it is put down?

better safe than sorry especially for newbies, imho.

I see your point but people really shouldn't be laying the needles down in between. If they do, they should be replacing the needle guard.

I reccomend the innoculation method explained on page 49 of Stamet's 'Mushroom Cultivator'. He suggests pre-loosening the lid of each jar and then moving from jar to jar innoculating and replacing the lids. Tighten the lids at the end. This really speeds up the process and minimizes the risk of any contamination.

i certainly don't agree that removing your lids to inoc reduces the risk of contamination in comparison to injecting through tape-covered holes. but whatever floats your boat.  :grin:

What? WTF does it matter. laying it down or waving it in the air. I'd rather lay it down then handle jars with the damn syringe in my hand. When your laying it down, its not like the needle is touching anything.. unless your laying it on carpet maybe. Your more likely to end up having the needle touch something when its in ur hand compared to when its not moving and just sitting there.

And whats with the problem of swiping the damn needle with an alcohol soaked paper towel. Seriously, it takes like half a second. I wipe mine at least once between jars, this way I dont risk cross contaminating, or risk having picked something up from the air while 'twirling' the syringe to redistribute the spores. Twirling isnt the right word, but I cant think of any other way to discribe it.

Wow, thanks everyone. I really appreciate all of the answers. By the info i have read this is going to be my method. Burn alcohol flame till needle is red hot then wipe it down w/ alcohol paper towel after it cools. Never dreamed you would have to be so sterile.
