Mycobag Colonization Log (6 days and no sign)

SWIM obtained a rye-based mycobag from mycofactory.com. The inoculation was done in a shower that was Lysol'ed down and the injection site was wiped with 70% rubbing alcohol. The spores used were B+ obtained from spores101.com. A bic lighter (yes, not the most ideal) was used to heat the needle followed by a wiping down of the needle with an alcohol soaked paper towel. An undesirable soot remained on the needle, but SWIM decided that it wouldn't be of much concern, considering that the needle was heated red-hot and wiped down with alcohol.
The needle was inserted and approximately 2 cc's were dispersed to the left, middle, and right. It was removed as the self-healing injector site healed itself.
After inoculation, SWIM realized that the syringe was not shaken, but it was handled and flipped a lot before the inoculation during examination of spores under a microscope, the removal of the syringe cap and the placement of the syringe needle, and during the inoculation process.
For the past 6 days, the bag has been kept in a dark box with an ambient temperature range of approximately 82-84 degrees. It now shows no sign of colonization, and there is slight concern. SWIM is considering a reinoculation. Are there any suggestions or comments to be made about this predicament?

I'd give it a little more time, and if possible, you might consider a bump in temperature by about 2 degrees. I had a similar problem as you did, and although I did eventually reinoculate, it was probably unnecessary, because once I stuck the bag into an 86 degree incubator the thing went nuts.

Ive got mine in a 85 degree incubator and it still takes a week before I see ANYTHING. Now if at 7 whole days I got nothing, I reinnoculate with the rest and THEN it takes off for the other week. By the end it's usually full colonized and ready to case.

Im on a bag grow right now and it's on day 5, so I'm waiting until Sun before determining if I will shoot the rest in there. I also use 5cc solution and then inject the other 5cc. I know it says 2 but if I use 2.5cc in the front of the bag and another 2.5 cc in the back I'm sure it will distribute enough in the rather nice sized bag.

Oh yeh, I don't flame the tip at all. I only take a cotton ball soaked in rubbing alcohol and wipe it all the way down. THAT is the purpose of the alcohol and the flame is a unecessary step that is built in redundancy that an engineer would like.

Thanks for the replies, SWIM will probably wait until Saturday before trying an reinoculation. Does anyone though think that the lack of shaking the syringe immediately prior to inoculation had anything to do with it? Note that the syringe was handled plenty beforehand.

Well, if even a small amount of viable spores made their way into the substrate, they should germinate. Of course, the spores tend to clump together in the syringe, so it's hard to say. I'd say you shouldn't worry about reinoculation until you've tried modifying a few other variables. Shake the bag up, break up any compacted parts. Try to get it around 85 or 86 degrees.

But I'm a newb, what do I know.
