OK, I have the manual, and ive read the tek. Just looking for any tips adjustments peolpe have used with good results.

The tek is basically cook a test tube of distilled water(pc) and scrape a wee bit of mycelium into the water. Close the tube and store at coolish temperature, but not in the refrigerator.

This tek is interesting, because there is no nutrition for the culture. It just floats there, and is reported to keep for many many months this way. Anyone have a tip, or used this?

Hmmm....months? Why not just use slants?

Distilled water is known to keep cultures viable for much longer than agar slants, owing to the lack of nutrients (the presence of which will keep metabolism going, resulting in the accumultaion of toxic wastes.) This method seems to work well for most species, except Pleurotis, which drowns in it.

The new edition of volume 2 of the peroxide manual (which all good cultivators should buy, well worth the $15) details another method similar to the distilled water tek that seems like a good idea: storing cultures in test tubes on moist, sterile paper pellets, with minimal or no added nutrients (you could moisten it with a very dilute malt solution, to give the culture some initial nutrition to speed colonization.) This method would be likely to work for most species, including Pleurotis, and would be easier to work with when it comes time to remove the culture form storage.

Hmmm...I don't know about that. Stamets mentions slants that were stored for 27 years and were still viable.

For cubensis agar might be fine, but for those of us who keep many species in culture, it doesn't hurt to hedge your bets. I personally use slants and H2O, and will use the paper pellet tek too.
