micron filters
    #448837 -

how do you innoculate a qt. jar with a syringe using one of these??? do you just use the filter with a band and no metal lid, or do you drill a hole in the lid?
do you push the needle through the filter??? or lift off the lid?
obviously i've never used these, but am thinking about purchasing.

thanx.

Sterilize the jars with the Filter disks on them, then after the jars have cooled get another filter disk and keep it close, Take off the band and lid but keep the filter disk on by gripping it around the neck of the jar, Innoculate the jars by sticking the needle through the filter. Cover the innoculation points quickly with the other filter disk, and wipe down with rubbing alcohol. Replace band and lid. After the jars are 50 colonized or so, take off the lid so there is air exchange.
You can leave the lid off the whold time but the jars tend to dry out, even if your incubation chamber is humid.
With a quart size jar I have found that if you just stick the needle in the middle of the filter and innoculate by pointing the stream of innoculant towards the side of the jar and do a sweeping motion, you get faster colonization and you only have one hole so there is less chance of contamination during the innoculaton. When you do it dont even stick it in the substrate and make sure you sterilize the needle first too, duh. I must say though that these filters are worth the price, I got the 100 disk deal, and have done alot of jars and made other things with them and I still have tons, and haven't had any contams in any of my jars yet.

Or if you already have metal lids you can inoculate as normal leave the lid on loose so they can breath and then when you have good colinization starting replace the metal lid with the filter disc for faster growth.
