question for workman on panaeolus substrate bag inoculation
    #3904454 -

Was wondering if you would be able to kindly answer this question...in your well done post on the panaeolus cyanescens grow, would it be possible to inject the bag with say a full 10cc spore filled syringe instead of a dilute malt water bath?

Workman's link:
http://www.shroomery.org/forums/showflat.php/Cat/0/Number/3751402/an/0/page/1

This would essentially make the process very easy for those willing to expend a whole syringe on one bag, just looking for a way to possibly cut costs and steps, since no laminar flow hood would ever be needed (in the malt water preperation? Just how is malt water prepared, is a flow hood essential, or are extreme sterile conditions needed?) and no need to grow out spores on grains, just inject the port on the bag containing the manure substrate or a substrate combo of manure, vermiculite, and ground brown rice or ground rye grain (1/2 pf tek, 1/2 manure combo in the bag, ie. for each 6 cups of pf tek, add 6 cups of manure) Even though you had success with just the manure substrate (no ground grains in it).

Just curious to hear yours or anyone's thoughts on this.

BR had mentioned in another forum that he had sucess with his formula in 1/2 pints, as well as full quart jars, and that the panaeolus mixture he uses even works in bags. It would seem pretty easy to just inoculate through the port with a spore syringe...everything staying sterile and such, no need to open bag in front of a flow hood for example to pour in grains, etc.) Just inject the presealed, sterilized bag after it has cooled off with a full syringe, shaking it up as you inject to spread the pan cyan spores all over the substrate in the bag. Don't know if this would work though.

Bob Robert's link to a cubensis grow with jars and bags that contain BJ's panaeolus mix:
http://mushroom.dk/svampe/cubensis/index.htm

Quote from Bob Robert's on growing pans using BJ's mix:

"Quarts only take about 2 weeks to colonize. You shake them up when they are about 20% colonized. You'll get some larger pieces that come to the top, but just make sure to shake it well and that you get some mycelium on the bottom. Jars are filled about 2/3. This recipe also works well in bags. The good results come because it's really pretty easy to grow them if you have your water content correct and you shake well."

BR mentioned that a quart jar full of mix colonizes fully in 2 weeks, just shake it once at 20% colonization, if this is true, then a bag of the mix inoculated with one full syringe should colonize quickly also???

How and why is it possible to inject pan cyan spores into mostly bulk substrate with or without some ground up grains, and watch it take off and grow so fast? Doesn't this break the rule of "don't inject your substrate directly with spores?" But then again, you used a malt water bath...looking for any and all help answering this question.

It would probably work but it isn't something I am going to test. I prefer the chunky broth method. Pan spores germinate quickly and grow well on pure manure in nature so my guess is that it would work fairly well in the lab. If the syringe is clean it should work almost as well as a mycelial syringe. It would just take a few extra days for the spores to germinate before mycelium is visible. Hope that helps.
