i have some isolated agar plates from MSS, that I have now colonized on grain, but I have never seen any of it fruited. this seems like the way to test the different strains before I G2G them into 10 quarts.

What temperature is your room? High 60s - 70s or low 70s - low 80s?

Starting my gallon zips. So I can test through some of these mono cultures I have made.

I sealed off the 2 corners.

I filled it with 2 quarts of water which would be the volume of 1.5q sub and a pint of myco spawn.

It’s basically up to the level of the label which doesn’t seem like it will leave much room for growth I saw you sad pop another zip on top. Does this look right though.

I put the water in so I could know where to start poking the holes in.

That’s about the amount of spawn and sub I use for my pans trays and I think they would work in a gallon zip.  You also need to take into account a couple things when comparing water volume and spawn and sub volume… the spawn/sub volume will shrink when mixed together because there’s gaps between the spawn that the sub will fit into and we don’t use whole pints and quarts etc for our spawn because we need to shake it so you can’t really compare the two equally.  I think you’ll be fine

And it’s not a big deal if you don’t get the hole pattern exact at the sub surface.. you can always add more holes once the sub is in there so I’d err on the side of too high rather than too low

thank you so much for getting back to me. i was thinking the water was going to be fuller than it will be in actuality. I stupidly filled my spawn jars to a variety of different levels. some are 3/4 but some are 1/2. the 1/2 full ones are basically a full pint. I am going to standardize how full I fill the jars on the next runs.

I think ill fill one up and then use that as a template for making the rest of them. the coir is cooling. so pics coming soon.

Welp I do love this tek, but idk if I did something wrong. its been about 15 day since S2B. All the bags are 2.5 gal bags with the corners sealed off to create a flat square.

I have not added any extra holes as mentioned is sometimes necessary. I used a needle from an old spore syringe, so maybe the holes are not quite big enough.

I keep the RH at ~55%. I have opened them a few times when I had to move them, and to help get the bag to stand up nice and tall.

one factor I am just now realizing could play a huge factor is that the MSS where inherited, definitely were at least a couple years old, and had been stored in various closets fridges etc. So maybe the genetics themselves are just pretty worn out. Should I try to just clone what grows and go from there, or try again, or source some new fresher genetics?

thanks for everything.

Nothing to really do but wait it out.  A couple look like they’re ready to start fruiting.  2 weeks is usually when I start looking for signs of fruiting so I’d say you’re still on track.  Depending on the culture, I’ve had bags/tubs take 3+ weeks to start knotting up.  Some of those do look like there wasn’t great growth from the get go.. that could be shitty genetics or dirty cultures.

Holy jeebus MB.  Very nice.

If anything, I have to add fae holes and/or open one side of the bag mid fruiting.

yup it was a mix of many factors. super glad to find this technique. Indeed a mix of genetics from older MSS agar plates. They are my first go around so im really glad they came around. I am pretty happy to get what I think are some pretty promising fruits for the next batches. Also these jars were both individual strain isolations, and also the jars that took the longest to colonize anyways, or where slightly questionable spawn. so it makes sense they were not great.

?Also I had a large number of side pins, I am going to look around for some rubberbands that will fit around these 2.5 gal bags, the ones I had didnt fit and I said f it. going to look for some tupperware, or rubber bands.

Mars (Albino) looks like I isolated out an albino strain very excited about this one. looks pretty similar to some APE's I saw recently

Mars these both came from the same MSS agar plate. both nice and dense. excited to get a nice clone of albino and non albino and do a tub.

orissa indica these end up being small, but I think the yield is actually quite good.

orissa indice as well going to look for the densest one to clone, but already a pretty decent canopy.

JEdi Mind fuck. these are nice and dense, definatly looking forward to a solid canopy of this strain.

also JMF not nearly as big, but nice canopys.

I have a Thai (Pink) Buffalo MSS.

I was thinking to just streak a plate or two, then if they are clean I can just let them grow out,

split each plate into 4 quarts

Then do each myco quart in a ziplock

select the best fruits to clone.

I figure this way I am getting access to all of the potential genetics, instead of if I take a few isolates from each plate.

If there is a better way to find the best genetics from a MSS please help me.

thank you

Surface conditions and health of fruits.  If they look like they’re stretching for air.  Usually it’s a good idea to increase fae mid fruiting in whatever container you’re using.  The fruits themselves displace air inside the container so that gets smaller and the substrate is pumping all its water into those fruits that are evaporating that water off their surface.  If you don’t increase fae, the sub keeps pumping that water into the fruits even when they aren’t getting enough fae to evaporate that water off their surface and the fruits become waterlogged and you’ll see faster degrading of those fruits; rotting before they’re meant to.
