Contam on my cloned lionsmane dishes

Heyhey,

so I just started growing mushrooms some time ago so please don't judge to hard if I missed something crucial. After trying to get into mycology for quite some time now I thought it was time to try getting into agar. One friend of mine had bought a lionsmane growbox online and I thought cloning these would be a good idea to first use agar. After I did the transfers though all of my dishes are contaminated now. What I don't understand is that for all of the dishes the contam is growing out of the tissue.
My ideas on reasons for this are:
There was contam already in the tissue used for cloning (my friend also spotted some mold in his box a week later)
After flame sterilising my pincers they had carbon grime on them because I used a normal lighter. Maybe that caused contam?

Apart from that I worked in a SAB and only took parts from inside the mushroom, which I broke open. It was all the same mushroom though since my friend only gave me one.

I would love to understand the reason for this contam and if possible also get some advice on how I could clone the mushroom instead (maybe taking mycelium instead of tissue?..).

Also excuse the poor quality of my phone camera. Also I hope I chose the right way to upload the picture. Still kind of confused on how that works..

Welcome to the Shroomery friend! :smile::mushroom2: glad you are here. It wasn't the soot on your blade, mushrooms are dirty so contaminations is common in clones. What Tek did you use to make your agar? In all likelihood you just need to work on your technique.
1. Plates are a no fly zone, don't let your hands or arms go over open plates
2. Long sleeves are a no go with SAB even rolled up they will wreck your plates.
3. Use one hand to open a plate from the back. Then quickly drop in your mycelium with the scalpel
4. Spray your scalpel with alcohol, and torch the blade and 3 inches back from the blade. Contam will hang out on the handle/neck of the scalpel.
5. Make sure there are no fans or drafts in the room you work.
6. Work quickly, time is of the essence, every second the plate is open the more likely it is to get contaminated.

I'd imagine you've read up on sterile technique, though if you haven't I strongly suggest watching a video on it. Makes all the difference. Best of luck and I hope this helps! Keep us posted if you have anymore questions:smile: below is a link to video with lots of info on the subject

I'm not all that familiar with all of the existent teks and the terms used and part of my knowledge also comes from youtube (primarily philly golden teacher, don't know if he is deemed credible here on the shroomery) and not just here. So for agar I just put some water with agar in an aluminium covered jar and the dishes inside my PC and poured it later in my SAB.

Thanks for the suggestions on cleanliness. I pretty muched payed attention to all the steps you mentioned but just used tweezers (confused the english word with pincers in the first post, sorry haha) to rip out tissue from the middle of the mushroom instead of a scalpel.

What confused me was that the contam was mainly growing right out of the tissue and not just at random spots on the dish like on the middle and bottom left dishes. I thought that meant the issue was not the cleanliness but maybe the tissue instead. Obviously that might just be a misconception on my part.

YouTube has some alright tutorials, I'm not familiar with philly golden teacher. Though I will say, you are in the right place now. Shroomery is thee place for cultivation info.

It makes sense that lions mane would be super dirty to clean up considering the way it grows a million little tentacles that likely capture a tone of stuff from the air. Kinda hard to get a clean chunk.

As you continue taking transfers to clean up your plates,keep in my mind lionsmane is very wispy mycelium, often fruiting before full colonization. It can handle it's fruits being chopped up (a couple times) the  sent to bulk. Inoculating with a prepared lions mane LC or LI and shaking will allow you to cover more grain before sending to bulk and minimize fruiting before hand.

Heres a photo of the inside on a lions mane fruit to illustrate how contamination is trapped within the fruit while growing.
