I could really use some advice if anyone can help. Im really new to agar and growing in general. The first plate was from spores. I thought it looked great for awhile, but now its getting darker? It was pure white a couple days ago. I know the spots are bacteria, but I thought i transferred from a clean spot. The 3 t1s looked good i think. My t2s are crazy tomentose, but good? It also seemed like it stunted the growth on my parent plate for my t2s when i did the transfer, although now it seems fine. Sorry, I know thats a lot. I did my best to take good pics, still figuring that out
Edited to say pics are out of order. Hopefully it still makes sense. I did not post the donor that slowed down, I posted a plate i think is clean that looked just like the one i transferred from. Sorry about the wall of text

Curious ... any advice on selecting an edge for transfer, or just feel the force and let the fingers decide?

You always want to target the healthiest, fastest growing mycelium. Doesnโ€™t matter if itโ€™s tomentose or rhizomorphic.

Hope that helps!

Anyone ever run a full 500ml of agar in a 500ml media bottle through a PC? 450ml works fine but 500ml seems awfully full. Donโ€™t understand the physics of it but they donโ€™t seem to boil over in the PC. Hoping someone has some experience with it though.

I use 500ml all the time in my 500ml media bottles. Never have any overflow issues. I just never fill above the little indentation where it starts to slope upwards to the neck. Leaves plenty of space for the agar to boil during the PC process.

Same is true when I use my 1,000ml media bottles.

It does, thank you.  This is my first germ plate, and it's just healthy uniform growth so far.  Will let it run a bit more to see if an overachieving section reveals itself.

If itโ€™s uniform across the plate itโ€™s most likely a monoculture, in which case you can take transfers from any of the edges. ๐Ÿ‘๐Ÿป

I use 400 just because I want that headroom if I have to microwave it back to liquid. Even at 400, there's only a few seconds to react before it boils over.

All the time.  I just loosen the cap a tiny bit and wrap the cap/neck with foil.

I do 500ml lid tight and water up to about half way up the media bottle in the pc

Which dish would you transfer from, and which section?

If you place the bottle in a hot water bath it will melt slowly and you wonโ€™t have to worry about boil over. It takes less than 10 minutes for a 500ml bottle to reconstitute in a stable temperature bath.
