mex-a....sectoring required?
    #1496119 -

How many transfers off the first plate (germination plate) are usually recommended? What type of mycelium is one looking for on the plate?
Does it matter much to go to like a second and possibly third plate transfer?
Or can the germination plate be used to innoc. the quart jars directly?

I usually use the first plate that is 100% clean. Since you'll probably want to keep the culture you'll make more transfers and try them later anyway.
I didn't try to germinated mexicana yet, so I don't know how mycellium behaves. I noticed sectoring on old mexicana B mycellium. Some sectors had fluffy appearance and I tried to avoid them.

Thanks for the reply.  Can anyone else confirm. How many petri transfers after germination are recommended before innoculating stone bags/jars?
Are you looking for rhizomorphs or a particular type of mycelium as preferable for stone formation?  or...as zeronio replied, you can simply use the first petri that the spores germinated on on.  -floydster

I usually transfer until there’s uniform growth of mycelium, making sure I’m not transferring contaminates from the ms plate. Once it looks more uniform growth I’ll wait until the plate colonized and transfer the quickest sector into grains or lc. Sometimes the mycelium will brown and will even make sclerotia on agar which makes great cloning

Typically takes like 3 to 5 transfers for me

Thank you both for the replies. How often does ATL7 form sclerotia on the agar? Is it usually necessary to do several transfers before she forms stones on the agar?

They will very likely grow stones on the agar, no required to transfer for it nessecarily.

I transfer twice from germination for a wide expression.

It's awesome you replied to this thread after so long.
