multispore mex-a on pda - many different sclerotia forming

hey!

so a couple months ago, i inoculated a pda plate with mex-a by streaking
some spores onto it, and as i looked at it yesterday, i recognised different sclerotia
forming on the dish.

the funny thing about it is, where the sclerotia should "meet" eachother,
they form distinctively seperated lines - why is that?

anyone with some info, please post, a pic will follow as soon as i can.

as u can see there are many different sclerotias and i wonder if it would be worthwile
isolating each of them and then starting to experiment with them individually ?

what u thinking of that?

Like a "No man's Land"? This could be the inhibition zone formed when two different substrains meet,there is an unclaimable territory.Maybe later on one will overtake the other.

Its quite interesting as a phenomenon.Thats the area where genetic material exchange of substrains can happen if some conditions are met (one easy is to utilize snake venom in the agar, look for posts on that)

Also ,could you post some pictures? Many of us i believe are dying to see it!

Thats normal, I think roger posted pics of it happening in the hybrid thread, he says once in a while a new line of growth will develop.

i will post pics asap, i wont open the dish unless opening in infront of a flowhood
for obvious reasons (dont wanna let that one go jabba jabba for sure !)

also, ill play around a bit with the dish - i plan on transferring each of the 3
sclerotia to other plates and test if they all got the same "potential"
concerning sclerotia formation or not.

ill keep you dated.

thanks, i already knew that thread.

this is about what my sclerotia dish is looking like,
i do not expect any culture to "overtake" the otherone,
i just think of it as being funny that severl different substrains actually
could grow and well uhm, live in co-existance to each other :wink:

I'm not quite sure what you're asking, but with multispore inoculation you'll get lots of different substrains forming, some better sclerotia producers than others. I'd transfer sclerotia or mycelium from the best producing sectors to either agar or grains to grow out. It's what I did with these plates. The best sectors were transferred, the rest were tossed out.

Multispore inoculation will result in MANY substrains forming, as many as compatible monokaryons will permit. These MANY will interact with eachother. Some will fuse together, others will be unable to.

The result is what you see. Several INDIVIDUAL substrains occuping their own section of the habitat.

It is definetly worth the effort to isolate as many substrains as possible to test for Sclerotia production invitro in your particulars, or fruit production in your particulars. You will find some substrains outperform others in your particulars.

Note. I find that even very productive clones(substrains) that have been tested, can still perform inconsistently JAR to JAR, casing to casing. But ALL substrains are like that, and it is better to find the ones that WHEN EVERYHTING GOES ABSOLUTELY PERFECT, PRODUCE THE BEST, amongst the many you tested. It may not be the bomb every time, but it is the BEST you found, when it does perform.

Roger, do you reuse your plastic agar plates or do you buy new ones? Or do you use glass pyrex plates?

Just thought I'd pick the mind of a mushroom growing genius. I've followed your posts for the past couple years and must say I have yet to be dissapointed.

they come sterile and dont cost a fortune, so why using em again as u cant pc them.

Agreed, use them once and toss them. I hate glass petri dishes. It costs more to wash them than the disposable ones cost.

Ohm, you can get an awesome dorm type fridge at Sears for $150. Get the cheapie one without the freezer. Ones with a freezer go into auto-defrost mode once a day, which raises the temp to nearly 80F for a few minutes. That will cause condensation on your petri dishes.

hmm getting one without a freezer sounds like a good deal, as i am VERY limited concerning space.

only deal is, it would have to be in the sleeping room so it must be very quiet as i am
super nervours to sound when its sleeping time.

are those small ones quiet ?

I have the one from Sears, and it's in my bedroom. I never hear it, and I'm also a light sleeper. I ponied up a bit more dough and got the stainless steel model just cause it looks cool.
