Sterilising 60ml single use syringes?
    #25757388 -

I have some single use 60ml lure lock syringes that I want to use with liquid inoculate but am a little unclear on how best to sterilise them before using them to extract liquid inoculant through a SHIP.

Any advise is appreciated.

boil water for 10 or 15 mins, aspirate boiling water in and out of syringe 3 or 4 times, on final aspiration leave boiled water in syringe or squirt it out in prep for LI

Sterilize it in the pc, don’t use boiling water.

why, boiling water is perfectly fine.

Boiling water IS enough to sterilize a syringe.  Plastic does not harbor bacterial endospores, and fungi like trichoderma and cobweb are killed by temperatures far below boiling.

If you have a pc why not use it? It’s no doubt more successful than boiling water

Lol, no. Why would you fire up a PC when you can just boil water? A PC will not be any more successful....  Besides, why use a syringe for LI when you can just pour it into any receiving vessel the same as you would G2G.

Maybe that is something I need to consider, pour from the LI jar to the receiving jar,

Indeed, follow same steps as per G2G. Munchausen has a vid showing the process if you want to look it up.

Fill the syringes with distillate or bidistilate water, or tap if you don't care, fill the up, cover them up with aluminum foil and pc them standing up, I normally use 4 jars to keep a bunch  of syringes in place, pc for 5 min at 15 psi and theyre good to go

Or.... suck boiling water into it. Why do people insist on making things as difficult as possible? Filling a syringe with water and then running it under pressure will cause it to expel it's contents.

Endospores could be on anything no? Op didn’t state the cleanliness of said syringe. For all we know it could be dirty as hell. I lack to see how boiling water is as clean as something that has been ran through the pressure cooker... pressure cooker is better than boiling water in any situation. It’s like pasteurization vs. sterilization. Pasteurization leaves stuff behind while sterilization kills everything.

A cursory glance through the forum is often enough to answer most questions, and will prevent having to post duplicate information ad nauseum.

I mean the entire physics of what should happen would mean water comes out of a syringe every time, no matter the orientation.

It's a known problem in industry too. The even make cycles that compensate for the effect.

Although I really have no reason to even use syringes anymore, let alone sterilize one with water in it. Besides its very easy to just suck up and squirt out boiling water 3 times

Quote:
Steam-Air-Mix Cycle
Certain types of sterilization loads may require steam sterilization cycles that stray from the standard Gravity, Liquids, or Vacuum types.

Take, for example, sterilizing a liquid-filled syringe using one of the three standard cycles. Immediately a problem is presented here: As the syringe heats up, the liquid inside of the syringe expands, exerting pressure on the stopper that is supposed to hold the liquid inside. This is especially critical when the container is topped-off with liquid (no air inside). If air resides in the container alongside the liquid, that will increase in pressure as well. If the stopper is not able to resist those pressures, the stopper will be forced out of position and may even eject from the syringe body. Now, not only will the syringe be unusable, but whatever was inside of the syringe will have made a mess of the rest of the load and your autoclave chamber.

The obvious solution is to increase the autoclave chamber pressure in order to balance out the internal and external pressure of the syringe (or other sealed container). If the pressure outside of the syringe is equal to the pressure inside, then the stopper should not move. But how is this accomplished?

One way to increase autoclave chamber pressure is to simply inject more steam. Unfortunately, as the steam pressure rises, so does the temperature. When the temperature rises, the contents of the container will heat even more, causing the internal pressure to continue to rise as well.  A Steam-Air Mix Cycle solves this issue by injecting compressed air into the autoclave chamber during the sterilization phase in order to artificially raise the pressure. Steam continues to be injected into the chamber as needed in order to maintain the cycle’s temperature setting.

After the sterilization phase is complete, it is necessary to maintain an elevated chamber pressure during the exhaust phase in order to keep the syringe intact. However, as the temperature falls, the internal pressure of the container will begin to fall as well, potentially causing the reverse-effect where the external pressure from the chamber compromises the container. To solve this, the chamber pressure is slowly reduced as the temperature falls.

The Steam-Air Mix Cycle option requires you to have a compressed air supply, and it is recommended for applications where a liquid is being sterilized in a sealed container that may be affected by pressure imbalances.

If they are upright no water will be lost from the syringe, yea man whatever floats your boat, but I wouldn't call boiling water sterile.

Yeah, you're probably right. I don't see how things could have been made anymore clear, I honestly don't have a horse in this race so do whatever you like.

No it's not. But for syringes it's good enough
