ok, the first time that i did the karo tek it worked great.and now trying it for the second and third time i dont see any germination at all after about 3-4 days. i used the spores from the same print and did everything the same. what could ve caused this.

What strains? sometimes it seems that some strains won't germinate in LC...I don't know whats up with that.

how long do you pc the karo for and do you use a filter of sorts? And how long before you see mycilium?

sometimes..with the karo tek.it takes 5 to 7 days for the spores to germinate..and then once they start to germinate..its a beautiful thing...just give it time..and keep em at a warm temp...

do you use a filter patch or just concelled in a jar? and what ratio do you use?

I have never seen someone who sweats contams as much as you.

You can use a filter patch, it works fine, but it is not needed and therefore is just more work. If you only fill your LC container about 30% and leave the rest as air, you should be fine.

I was telling some one the other day how overboard a lot of people get with sterility!

But the filter is uneeded, extra work, and prone to contam, so don't use em.

Make sure to keep the karo/water ratio around 2tsp karo/1pint water. Anything more is probably too much. Thats the only other thing I can think of right now...

I did a half pint jar with 1/2 cup of water to half a tablespoon of karo and it filled the half pint jars abot half way maybe a little more is this bad or will it work even for a couple of syringes? oh and I boiled them for ten minutes should I pc for 20 minute isthis better?

I dont use any filters. Solid lid PC'ed for 20 minutes rubber side up and wrapped with foil. After cool I take to my glove box, remove the foil and take off the lid, shoot it and turn the lid over and seal it tight. The foil keeps the underside clean while in transit so that when i remove it in my glovebox i can flip the lid. I use 75cc water and 1 tsp karo.
Works good.

why dont you just make a hole in your lid prior to pc'ing and tape it up good then do the alcohole, flaming needle, inject through the hole and while pulling the needle out put another piece of tape on as your sliding the needle out then you dont need a glove box and all that hasle

Its not any hasle and I have my glove box so I use it. But there is anything wrong with a hole and tape.
