Morchella?
    #21437349 -

Hey all,

I've recently done some tissue isolations of morchella esculenta and I am wondering if any of you knew what M. esculenta looks like on a culture plate. I've found a lot of images of black morel, but yellow morel seems to be harder to find. The mycelium is somewhat clumpy like images i've seen, but it has lots of dark spots which I have not been able to find a comparison to. Possibly sclerotia? If anyone could give me some advice that would be great! Thanks!

Awful lot of green in those images...

Here is what mine looks like, bear in mind that Morchella is a diverse species complex, not all M.escelenta is the same species so appearances will vary wildly. Mine was collected from Upstate New York and I think is most appropriately called Morchella americana, though I have not read the recent studies on the Morchella genus to see if I am correct.

The sclerotia does not always form, but it seems to consistently turn the agar underneath a dark reddish brown color.

If you have access to a microscope I would try seeing if that green is spores from some mold or just the coloration of the mycelium.

Hmmm... Yeah, this is a variety I found in central california. I also asked a mycologist at the local university and she told me that since the spots are forming uniformly across the plate rather than localized to certain areas it's most likely sclerotia. But you are right, too green for my liking... Though, I decided to transfer one to grain to see what happens.

Sclerotia are never "green". The sclerotia of M. esculenta is always from white (when first developing) to reddish/orangish (with age).
And since you are in California it is unlikely you have the "Big Blonde" fried chicken looking M. esculenta species of the midwest.
Its more than likely a Pacific coast blonde with lateral pits aka M. frustrata.
M. esculenta tend to look more like an irregular club shape sometimes rather than a Christmas tree.

Compare and Contrast Morchella frustrata and Morchella esculentoides.

What's happening to your morel culture is this: it is growing but it is being attacked by a predatory green mold and because it is being attacked it is producing micro-sclerotia although hopeless. Find an uncontaminated section and get sample, re-inoculate another petri dish.

Good to know! Thank you. I will try to section off a clean piece right away. I guess the grain spawn won't be any good but I will grow it out anyways just to see what comes of it. Also, i think you are right. It is M. frustrata not esculenta. My guide book didn't have M. frustrata and I chose the closest looking one. These are what I collected :
