will bottled H20 instead of distilled H20 effect my PDA ?

i dont have distilled water in the house, only bottled water..is it ok for me to use that for my PDA mix instead ?  will it even make a diff ?

professor pinhead: can i pour the agar in my plates before i pc ? i was planning to wrap them in foil and have them lifted on a rack in the pc? so i dont have to pour my plates after i pc..all i gotta do is let them cool, then inoculate with spores.  is there somethin wrong with doin it like that ?

What are you using for plates? If they are the disposable ones they are probably not autoclavable. If you are using jars you can pre-pour. Otherwise just pc it in a quart jar, then pour right after the PC loses pressure while its still hot.

If you don't have a flow hood clean the shit out of your bathroom before doing this and use a glovebox,.

Then You just inoculate your agar the next day when it has cooled,. Again in a glovebox.

I never worked with agar until I had a flowhood though... People didn't really do that when I was learning. It was a big no no....

ive only got 2 petri dishes that i can use right now, and they are made of glass.. so, it should be ok right?  and i should wrap them in foil to keep water from getting in right?  ok, will do.. ive gotta glovebox i can use.. i was gonna use the shmuv box tek, but i dont think its worth it,,  pure still air in the gb is just as effective as running a cheap wal-mart hepa imo.. im only gonna make these two plates, cause its all i really need..after i get a good isolate, im gonna eventually just use g2g to keep it goin..

I just poured some Ager in my Glove box. And all 20 plates turned out good.
I inoculated them in my glove box as well. And they are good. But word of caution.
It is a pain in the ass to do all that work in the confines of a glove box.

I am in the process of building a Flowhood as we speak and I will say this.
With all the work it takes to build a Flowhood. You should build a flowhood.
This think is simple to build. It is not much harder than building a glove box IMO...
A little more $$$ but if you follow stonesuns flowhood build it will run you less than $300.00

I just proved that. Because I just purchased everything on his like and it was less than $300.00
But... If your in this hobby and cannot afford $300.00 within a decent amount of time. you may want to reconsider this hobby :laugh2:
I easily already spent a couple grand.

This is what I recommend you do.

Glass petri dishes are a pain in the ass to pour due to there weight and when you got alcohol soaked gloves
You are GOING to drop one. I promise you will drop one... I have dropped them... I remember RR saying he dropped a shit ton...


So... because they are glass. I say you should pre pour them...

Mix up you agar. pour it into the glass dishes. PC The pored dishes with the agar.
wait until tomorrow to take them out, so that have hardened. put them straight into a cleaned GB
and then do your thing.

i hear ya bro.. ive been wanting to build one just like stonesuns.. but, for the past 4-5 months i havent been doin any growin, so im just now setting up shop again.. and ive had great success with my glovebox..and i know what you mean by it being a pain in the ass. limited room sucks... but, i need to make a flowhood anyways, so i can have it on a timer in my growroom to scrub the air every 2 hours. and to work infront of it aswell.. ive invested so much money and time into this hobby too, i know what you mean bro..  thanx for the advice..
  i will do that now actually.. ill keep you posted..  o yea, do i cover the plates with foil while i pc them ?

They'll boil over.  Sterilize the agar in a bottle or jar and pour later.  You can sterilize the empty glass petri dishes at the same time by wrapping in foil as you said. 

Tap water is fine to use.  If you have a filter, use it.

Didn't think about that they would boil over... I used jars when I pre poured.
Your not going to like pouring the agar into the glass petri's tho... it sucks.

I am so excited I am building a flowhood. no more worry about this kind of stuff.

If you can control the PC you can prevent your plates from over boiling. It may be a PITA but it can be done - I've done it.

I don't like to have to fire up the big PC so I sometimes pre-pour five plates (small stovetop PC), wrap them in foil and PC them on the stove. If you pour them thin, keep them level and PC them less aggressively it'll work. You'll have to bring the PC up to pressure slowly and try to keep the PC from releasing pressure during the run because every time the rocker starts moving and letting pressure out the agar starts boiling (releasing gasses - like a scuba diver surfacing too quickly). The boiling may even start when the pressure cooker starts cooling down too quickly - try to decrease the heat and let the PC come down slowly. Slow ride up, slow ride down. It's really not that hard, you just have to control it. Turn it off before you go to sleep - they should be solid in the morning.

I've been curious about this lately and will try it tonight or tomorrow. I have a different type of PC (counter top) - it doesn't have a weight that moves, it's heavily insulated and it is computer controlled. I have a feeling that I can put in my pre-poured plates, punch in 45 min and walk away. It heats up, starts the timer and once finished goes into a 'keep warm' state that brings it down slowly.

Give it a try - you may end up with a couple of plates that are pre-poured and condensation free.

o, ok.. well,, i already pressure cooked the pda in a pint jar, ive got the 2 plates wrapped in foil, and 4 empty jars in the pc aswell..ill pour the left over agar in the other jars..im cleaning and getting my work area clean so i can pour this shit before it hardens..

well, im all done pouring my plates.. all is good.. i had just a little bit of condensation building up, but not much.. i filled a glass up with hot water and set it on the top petri dish to keep some heat on it.. tomorrow i will scrape some gt spores in there, and see if all goes well.. ive got 3 jars and 2 plates with pda..i hope all of them are sterile.. thanx again to everyone that helped me answer some questions..

Sounds more like a light weight autoclave than a PC. .....
