The pickup closest to the bridge is your rhythm pickup. The one at the top closest to the neck is your lead pickup. Rhythm is meant to be played for rhythm parts, lead for lead parts. The lead setting will have a brighter tone than the rhythm, thats how you can pick them apart if you have no label.

Depends, what is your kind of Guitar?

H/H ? (Mostly Gibson)
S/S/S ? (Strat Style)
H/S/H ? (Ibanez)
S/H/H ? (Uuuh...)

Let's say, for a H/H guitar.
Les Pauls have it this way:
A switch for Treble and Rythm.
The pick up that has the ''treble sound'' is the bridge pickup.
The pick up that has the ''Rythm sound'' is the neck pickup.
This is for every guitar... the warmer, softer, ''muted'' sounds  come from neck pick ups, while the dry, harsh, open sounds are from the bridge pick up.

You can choose to have the switch in middle position, between the 2 pickups. The sound will be a combination of both.

S/S/S guitars will have 5 positions.

S / S / S
1 2 3 4 5

H/H, 3 positions
H / H
1 2 3

#1 is the closest pickup to the neck of your guitar.
#5 (or 3) is the closest pickup to the bridge of your guitar.

If you screw your pickup nearer to the strings, IF I REMEMBER, the sound will be clearer - but not better.
It's all about preferences.

Did I help you?
