Strain Isolation Methodology

Greetings. I have a simple quick question about cloning/strain isolation.

One of my books states in detail to clone using the tissue (mycelium) inside the stipe.

Then I have an older video reference telling me to split the cap open and that the best sample for tissue culture being the inner cap?

What one is the best? My gut tells me the stipe, due to actual visual mycelium growth.

If you have an opinion on your preferred method please share it.

All sterile techniques will be used, laminar flow hood, sterilizing the mushroom to be cloned via alcohol. ect ect...so don't bother with chatting about that. thanks.

The stipe seems most logical to me. For one it's just easier to work with, easy to handle, the outer tissue peels back to revel the inner tissue... second there is less change of accidentally "contaminating" your clone with spores from the cap.

Yeah my thoughts exactly, I was thinking the same thing...earlier in the video the guy explains IN DEPTH how spores spreading in the lab is horrible and bad talking about agar transfers and stuff after that...then he fucking tells you to take your tissue sample from the cap.

Contaminants are always more readily found outside and surrounding exterior tissue than they are inside an organism. So the most logical thing to do would be to take a core sample from the stipe.
