Cultivation of 7 strains in pasteurized straw

Sterilization of the straw-based substrate was replaced by pasteurization at 65C and 98% relative humidityfor 24 h. Inoculation was performed by mechanical mixing under conditions allowing natural contamination by spores of Trichoderma sp., as above. Blocks of 1 kg were formed and incubated at 25 ? 1?C with a12 h light/12 h dark cycle for 5 to 8 weeks. Four trials including 10 blocks for each strain were performed. One block per strain was sampled at spawning, after incubation for 1, 2 and 3 weeks and when the blocks were ready for fruit body production (5 to 8 weeks). To estimate the apparent level of contamination by Trichoderma sp., the area of zones colonized by Trichodermasp. and L. edodes were measured at the surface of the blocks using a graphic integrator. The mycelium of L.edodes was dense, whereas the mycelium of Trichodermasp. was diffuse and it was rapidly uniformly covered with typical green spores.
