APE Spores Not Germinating on Agar
    #29664226 -

Hey new grower. This is my second attempt at growing in years and it's not going well thus far. It's been 5 days since I inoculated 2 agar dishes with spores but I haven't seen any signs of growth whatsoever. No contam either. I plan on using these to make successive transfers until I have a clean culture at which point I will inoculate grain jars.

I bought a APE Spore Syringe from eden shrooms and I used the following technique to inoculate my agar:

1) Mix up syringe
2) Spray small amount on liquid in cup
3) Use sterile cotton swab to pick up liquid
4) Use Z motion to inoculate agar plates with swab

I recorded myself and included a video below (i couldn't figure out how to embed the video).
https://anonmp4.art/v/mngKCbybIA53DzP

The agar is 12g MLE, 10g Agar powder and 2g activated charcoal power.

Everything looked decent to me, you are being more careful than I am typically!

I have some experience with germing APE.

APE is pretty unreliable when it comes to spores. Not only does it produce way less of them (some individuals never do), but they have a slow and low germination rate. Luckily with fungus you only need two compatible spores! They can take much longer than 5 days though, so don't give up on them just yet. I've seen germination after almost 30 days in a dish I was is going to throw out but I'd been putting off.

I'd say wait and see first .

I am old school and opinionated on certain things. A couple of suggestions:

I often get pushback for this but I am of the opinion that agar should be inoculated from spores from a spore print primarily, or a heavily laden spore swab taken from a spore print. Also that a spore syringe, as was designed, is meant to inoculate BRF/verm.

What you could do is inject your spore solution into a BRF/verm puck either using the instructions in my sig (Combo tek) or similar method. You are more likely to get germination and you can then transfer the first puff of mycelium or a pin to agar then proceed normally.

And back to what you are currently doing, as tbone said: "APE is pretty unreliable when it comes to spores. Not only does it produce way less of them (some individuals never do), but they have a slow and low germination rate."

As he said 5 days is not enough so give it more time.

Also regarding your use of the swab. As you are lightly moving the wet swab across the agar very few spores will be transferred and they will be suspended on top of the agar in  a thin layer of water. Dig in harder and finally bury the head of the swab right into the agar. Then you know the spores will be in direct contact with the agar with a better chance of germination.

But I suggest the BRF. Worth a try, right?

By the way you have a great setup and technique. Good luck.

Personally i think the back and forth from syringe to swab after what sounds like a dilution might be an issue. When using spore syringes to agar, which I have done many times, I would melt a divot into the centre of the plate with a hot scalpel or loop. Then drop a single drop into the divot, this makes it easy to keep in one spot and prevent it from rolling around. Spore can take a long time on agar, especially if they are old. Probably shouldn't mention sponsor names here but I would go to the original developer of APE if I wanted to grow them.

APE are a bit finicky at the best of times and require a lot of patience. But they are also a lot of fun. Good luck

The only spores that have ever shown life in 5 days were taken from a very fresh print. It was less than a week old that I made myself.

I don't start to worry until at least 2 weeks, and even then I'll still wait because I have had some that take 3+ weeks to show life.

Be patient.
