the definitely not official fafo thread * 14

So this is a place to share our random experiments.

As an example, I've tried hydrating sclerotia jars I wasn't attached to but I didn't feel like I had a place to document that.  I believe the stones appreciated it but my grains were probably dry.  Might be fun to explore further.

Anyway, please share your thought process behind your experiments, I would like this to be a little more than throwing poo at a wall and hoping something sticks.

But experimentation is encouraged. Sharing your successes and failures is also encouraged.  We can all learn from both.

Currently, fafoing some nixtamalized popcorn.  My thought process is that hominy is more bioavailable for people than corn is.  And it breaks down mycotoxins that microbial fungi may have left on the grains.  That are a cancer factor for people.

I'm not sure how this really translates to mycelium, but I did see an old thread where someone was using canned hominy to grow mushrooms. And that stuffs full of salt, mine isn't, so I think it will have a pretty good chance of succeeding.  Don't know if it will be of a benefit or not.  But I'm curious

My process was to cover three cups of popcorn with reverse osmosis water, add a tablespoon of calcium hydroxide and simmer for 20 minutes.  Add more water if needed.  And then let the grains soak in the water overnight.

I then strained and rinsed the grains. Next time I think I would actually wash them in some cold water to remove all of the starch because they did stick quite a bit when they came out of the pressure cooker.  But they didn't really burst or clump.  No additional simmertime was needed to hydrate the grains which was really nice for popcorn.  As I usually have to cook it for 40 minutes in the instapot to hydrate.

Anyway, the jars were inoculated today with some pan mycelium.  I will report back in a couple of days and let everyone know how they look.

I messed up and accidentally threw my last good PE6 plate before making a transfer. I did have one that was poured so thin that it dried out completely and stuck to the bottom of the plate. As a last ditch hail Mary, I decided to try an inverse cabin sequester. I dropped a large square of agar on the dry mycelium to see if it will jump and give me something I can transfer.

Oh you know I'm on board for this!  My current FAFO is a wild oyster tissue sample in stasis.

I made clone plates as well but wound up throwing em out.  One never started growing out and the other looked like unformed teeth and eyeballs.  I should have taken pics lol.  Anyway in a few months I'm going to attempt to clean these up.  I'm trying to see if it's possible to keep clone tissue in stasis and check viability.  I'm sure it's been done, and I know there's a lot of cleaning I'll need to do.  I think this might come in handy if it works, let's find out!

So I had seen in the updated pf tek guide huskies said that you can use any flour. Either he or someone else said you can also use hydrated coir in place of verm. Made 5 half pints of coconut flour and coconut coir. Gonna noc em tonight. If they come out clean I'm gonna find a way to spawn them in coconut shells so it's 100% coconut except the myc.

The coir I can't vouch for but the different flours I can.  I've used rye, millet, spelt and WBS flours with success.  Definitely keep us posted!

You can colonize any substrate the pf way. Coir doesnt compress as much as verm. Iirc psilocybe fanaticus did all the work of seeing what works best before he settled on brf and verm

I'm definitely curious about the coconut flour.  Because most flour is made from grains and the nutrient composition in coconut meat is different.

Plenty of sugar though!  Looking forward to how it turns out.

Suggestion, little umbrellas for when you post them in the novelty thread

I have messed around with dried out agar plates and had some luck.  Had some ttbvi plates that had dried way out....Mycelium has long since reached edge of plate.  I had cut wedges in past for transfer....

I put a chunk of the dried out plate on a new agar plate and it took right off and pretty sure I ended up getting fruits out of that experiment.

Old dried out plates can very well still have life in them.    Shouldn't be surprising I guess.... Sourdough starter and yeast survives dessication.

Wouldn't be very FAFO or noobie of me not to try just cause someone else already figured out the optimal path now would it?

MiB recovering well on hominy popcorn.  I have four other pans on it.  And two of them have regular popcorn controls.  All are showing some signs of growth but this was the most photogenic.

I'll post an update before I shake.

Going to shake this morning, jars aren't photogenic.  But tropicalis and bisporus(LI) show minimal recovery.  Chlorocystis (LI), MiB, and a cyan marked red giant good recovery.

Bisporus and chlorocystis controls good recovery.

Planning to prep more corn today and run it tomorrow when it's cooler.  Have two different varieties I want to try.

Turns out mycelium doesn't really mind eating coconut flour! Or it's still pulling nutrients from the agar

Bonus FAFO: while pulling transfers from that Gandalf plate I had a slightly dubious spot that I hemmed and hawwed about. I had a control jar of grain nearby and the intrusive thoughts won.

This looks questionable at best

Awesome!  Looks happy and healthy too.

I'm prepping more hominy.  Blue and yellow popcorn, and controls of both.

20 minute simmer and overnight soak wasn't enough, so simmering another ten minutes, and then letting everything soak while my controls are hydrated in the IP.

So, 30 minute simmer and overnight soak next time.

edit:  chlorocystis, bisporus, and MiB showing good recovery after the shake.

I'm excited about the chlorocystis.  It seems to appreciate the hominy.  And is found where there's a lot of natural limestone.  So, not a surprising result but hopefully one that will lead to better pinsets.

Hominy on left, controls on right.

A couple of observations.  Washing the hominy didn't really improve the stickiness.

And hominy plumps up way less.  I used one cup of popcorn for each prep.  And got around 2 cups of hominy per cup and 4 cups of corn.

Also, I really like the baby blue popcorn.  It's holding its shape well and I think will be a bit easier to work with then the lady finger (small kernel yellow).  Plus it's high in antioxidants and other good stuff.  And a tad cheaper than ladyfinger.
