How do my jars look * 2

Obligatory "this is my first grow". I have spent the last few months lurking here and consuming as much information as possible while simultaneously experimenting with agar. I've now gotten to the point where I am getting pretty good cultures with a very low contamination rate using a homemade SAB. I prep my plates using Milkboy's PDA recipe.





After my last round of transfers I took the 4 best plates of GT I had and inoculated 10 jars of wheat (10/9). I prepped the wheat using Beef's Wheat Tek, and inoculated in my SAB using all of the usual sterile techniques. All 10 jars started colonizing quite quickly. When they were about 30% - ok, maybe I got a little ambitious and they were a little less than 30%? Like I said, this is my first time - I gave them all a good shake (10/13). After the shake the jars bounced back at different rates (expected). The first 4 were nearly fully colonized within 2-3 days of shaking, and the next 2 are just a few days behind. I had 2 that were clearly bacterial - confirmed when I went to toss them and they stunk like rotting garbage. There's one that, after looking at it today, I'm pretty sure is also bacterial - it will get tossed tomorrow. Then there's one that is suspect, but I'm letting it do it's thing in isolation in case it's just a little bit of a laggard. So I'm including pictures of what are, in my complete novice opinion, the most promising six jars from the lot.








The first few have developed those thick white blotches over the last 2 days or so. Is this something to be worried about? There are also a few uncolonized grains in the fourth jar - they're not completely uncolonized, but you can see that they're not completely colonized either. Jars 5 and 6 are the ones that are 1-2 days behind. Interested to hear everyone's thoughts, as I'm aware I don't really know what I'm doing.

I am currently soaking some grain for my next round of jars. Last time I took transfers I also made some LC. I tested the LC on agar with no contam, so these jars (and the next few round of jars after it) will be inoculated using the LC. Including pictures of those here as well - one is GT and one is APE

Plates look clean and apart from those blobs your jars seem ok, but, those blobs, wtf, they don’t seem like pins so I’m stumped, maybe just fruit them to a fahtlock bag, jars without blobs seem safe though, send them! πŸ‘

It kinda looks your jars are blobing as there pinning on grain. Might be bacteria from lc. Keep is posted.

Great. I appreciate the feedback. I have 20 other jars that I am about to inoculate with my LC so I am going to go ahead and spawn these 6 to bulk and see how everything goes. I will post an update once I have one.

Jars without the weird circle stuff look good, I would separate them and spawn them separately.

Side note, I've had mycelium do that before and Spawned it, did get fruits. I'd still separate them tho.

Appreciate the advice and the kind words. I did end up spawning the 4 "cleanest" looking jars to bulk on 10/22. They all smelled wonderful when I opened them - frankly I enjoyed that smell a lot more than I thought I would - and so far the tub seems to be colonizing nicely. I will post a picture at some point this weekend when I get a chance. I will likely spawn the other 3 jars with the blobs to a separate tub and see what happens - you know, in the name of science.

Separately, I inoculated 10 jars with the APE LC this evening, and plan to inoculate another 10 on Saturday. I may have gone a wee little bit overboard and bought a metric fuck ton of quart sized jars and two (ok maybe three) 50 pound bags of soft white wheat. So between all of that and the 4 jars of LC I made, I think I will have my hands full for the next couple of weeks.

Sorry I haven't updated in a few weeks, things have been crazy around here. After spawning the 4 best looking jars to bulk my monotub began to colonize nicely, but after a week or so more of those blobs started showing up. It was putting off a ton of metabolites, showing no signs of pinning, and eventually the tub started to smell, so I tossed it. Here are some pictures of the tub as it was colonizing, as well as right before I dumped it out in the back field.


on 10/29 I inoculated 20 more jars using the LC I made - 10 GT and 10 APE. The GT colonized pretty quickly and I shook them about a week after inoculating. Of the 10 GT, 3 are showing clear signs of bacterial contamination, but the other 7 fully colonized within a few days of the break and shake. However, these are now also developing the same blobs I had in the first batch. From what I read it could be stroma, but like I said I don't really have any experience with this so I am just going based on what I have seen online. The weird thing is that these blobs don't start to show up until the jars are fully colonized, and up to the point of full colonization everything looks like it's progressing well. Here are those jars for reference


The APE jars are, as expected, taking much longer to colonize. These aren't showing any signs of contamination or weird behavior so far, but neither did the other jars until they were fully colonized, so holding my breath as these continue to colonize. Here are some photos of those jars



Does anyone have any ideas what might be causing the blobs in the GT jars? I have now seen them in jars inoculated from two separate cultures. The temp in the closet I am using is always between 73-76 degrees and my jars have filter stickers on them to allow for sufficient FAE. I thought it might be related to the grain I am using - maybe some sort of bacterial contam that's surviving the PC cycle - so the 10 jars I PC'd yesterday were PC'd for 120 minutes rather than the usual 90. Would appreciate hearing from anyone who has dealt with something similar, as right now I can't seem to even confirm what the issue is.

Not sure if they bruise or not. Once I spawned the jars to the monotub I tried not to open it or touch the substrate. Once the tub started to smell I tossed it - again without touching the blobs. Maybe I will spawn the jars I currently have to another tub and see if whatever grows will bruise.

I'd take a step back and test my LC on agar before trying it out on more grain.
