Re: A strain crossing thread [Re: MycoBrainz]

I just started out reading up on crossing varieties and I feel this might be the well of knowledge I looked for.

I want to create a RW x NSS cross. Before I start... Has anyone already made that particular cross?

NOt many people have played with nss as a cross, Cronicr and Pasty have done some great work.

Tedsdead has also been busy, check out his Trump dicks.

Lol, Trump dicks!

I will certainly check them out. I STILL struggle to navigate here. I mainly use the search function but I am getting there.

Meanwhile I read through half of this thread. What a rabbit hole... My kind of rabbit hole.
I see there are many different ways to attempt a cross from mixing two grain jars with different varieties in one tub, over mixing liquid cultures, etc.
I got two prints on foil and want to place spores from both on one plate. What I gathered so far is that I have to place the two different kinds of spores as close as possible to each other on agar right?

I can't wait to do that. I actually see myself doing that this evening already, but I feel I need to read up more on this. What would be your take on it seeing that you are experienced with crossing? Jump into the deep end and learn along the way or read, read, read first?

When you search make sure to limit your search to the last 4-5 years.

If you have 2 separate prints, there's a few ways to cross...

My personal method is to swab print A and B on the same swab and streak plates with the mixed spores.

Others prefer to swab print A and print B separately and then make a cross hatch pattern going vertically with swab A and horizontally with swab B

Theres also others that use sterile water to help collect and hydrate the spores. They'll also swab or scrape spores into sterile water to create a spore syringe with the 2 varieties of spores together.

You have plenty of options, if it were me, I'd jump right in the deep end and start swabbing!

Happy crossing!!

Ah, that's a good idea DERRAYLD. Thanks!

I'll do it they way you do it, roarkell. Into the deep end it is. I got a feeling this is going to be even more fun than "just" growing.

Thanks again for the guidance, you guys rock.

Mix, streak, wait, select and transfer, test.

A recent flush. One of several Falbino (F+ x PF Albino) offshoots. Falbino 7 to be precise. 10 year old spores. I shelved it due to poor albino production. It still pops a few out from time to time, but it's rare compared to Falbino 3 and 5. Even so, it has meaty caps and solid stems with a great buzz.

Blowing the dust off this pic. A Falbino albino from one of the early experiments. This is from the same batch Workman used to make APE 2.0
Sure is fun watching you all doing this. Brings back some really good memories.

 Yeah something extra magical about that pure white.
Here's another chiseled from the sediment.

Once the universe comes into alignment I'll get back to work on improving a pure albino strain. I have several projects that were shelved thanks to zapos, vivas and life in general. Most of what I've been testing is still viable after a decade. The next test will be one of the first back crosses. It was doing some interesting things before I got side tracked.

This one was so big it sucked the cake blue.

Solid stems too.

The chosen one! These are some beautiful fruits. These are so white, they look like they come straight from a washing powder advertisement.

Few months back i had this plate that grew with much more of a deformity than previous grows, from the same spore print. Am I wrong to think that this looks exactly like the F2 generation in the op? second photo is how they usually come out, i actually saved this strain from a tiny, piece in a dirty bag of the first powerful mushrooms I'd ever gotten my hands on prior to ever even having grown mushrooms, with the thought of having a professional bring it back for me - I ended up throwing it onto agar myself after getting into it

Those are chunky.

You make prints? How long ago did you get the originals?

Unfortunately I chose not to at the time due to lack of supplies, and the general fact they weren’t producing spores. I know i should have attemped to, but i do still have the mushrooms themselves and the spore prints i grew them from. I got this strain by simply using a scalpel on a completely dehydrated piece of a cap that was probably grown by the original person around april-may 2020, i got the strain onto agar sometime later in the fall, photos of these were about 3 months ago

I don’t see why i cant repeat the same thing with the remaining physical ones - even though they’ve been thoroughly dehydrated
Still inexperienced, would love ideas on what I should do next with this

Recapture it again and then keep a culture going on agar...

Re: A strain crossing thread [Re: Nichrome]

2 weeks ago I had a shoebox containing MS TAT and  possible "revert" clone Melmak grain spawn. The first flush consisted only of TAT. I then prepared an outdoor patch using this substrate and this week a variety of pins, including I believe two albino phenotypes and two non-albino phenotypes, have popped out:

The brown-red pins closely resemble the color of the caps produced by the Melmak culture:

(From a separate tub)

Leading me to think that the more orange-capped ones might be a cross especially  considering the Melmak culture was based on a clone of a plate pin.

I've left a few of these orange pins in the patch but removed a section of the substrate from which two were growing and placed it in a jar to hopefully allow them to mature a bit more in safety until I can swab some spores:

These mushrooms exhibit the dimpled appearance of the TAT phenotype where the cap connects to the stipe:

(Same TAT culture from a different tub)

Still possible it is not a cross at all of course. If I cannot obtain spores I will make a clone and swab the resultant output.

That's pretty sweet, I've seen some weird shit in my spent substrate pile outside as well , your post makes me want to experiment

Update on the suspected TAT × Mel. This cap morphology is really convincing me that this is indeed a cross as it is very Ghostly.
