Unidentifiable problems, please help
    #25559586 -

Hello Shroomery

My account is new although I am not. I have successfully used Agar, LC  grain spawn etc everything for 6 years. Have a strain I've isolated and been using through LC and then G2G for new Jars

My problem is that now, all of a sudden nothing is working and 8/10 Jars/grains/ and cakes are all contaminates? ...

Exact same methods always used, same equipment etcetc started again from scratch and did things differently to work out variables.
I have literally tried and changed everything and am stuck needing help, hence this post.

I have very good knowledge and experience with shrooms so obviously I wouldn't post without having a read through and knowing everything I'm doing (except that 1%) is on point as usual.

Jars are not kept to hot when colonising
Water/Agar/ Jars and grains are all 100% cooled down enough so spores and culture are not being vaporised
Everything is done in a fully sealed glovebox navigated by me, OCD clean freak isopropyl spraying Nazi.
All scalpels and needles/plungers are open fresh sterile packets and flame sterilises

Seriously wizards, help me I've been loosing some serious shit..

Thankyou for your time ✌️

You've got to be able to work this out somehow.  There must be a simple explanation.

Could the strain have become contaminated?  Perhaps you could start from spores again to test that?

PC some jars and don't open 'em, just incubate 'em to see if they are getting indeed getting sterilized.  Maybe it is your PC.  Its gotta be something.

Maybe your filter disks are shot?  They do wear out eventually I hear.  Maybe try some polyfill for a few.

There's another thread from today where people are talking about needing 2.5 hour PC cycles or taping quarters on the weight to make it go to 20PSI...

Did you try that?

Use controls. Lots of controls. With every stage of your work, leave a few containers uninoculated. Go through the motions of inoculating them, except don't actually inoculate them. Like for an A2A, flame your scalpel, cool it, pretend to cut out from your donor plate, place the non-existent tissue in your receiver plate, close up your plate, and seal it as you normally would. Do a bunch of controls at different stages. There should not be anything growing on them after some time. How long is some time? I'd say at least double your normal grow time for that stage.

I had  what appears to be a bacterial problem stemming from my grains that weren't sterilized properly due to my altitude. The bacteria has not yet shown itself in my controls, but the strong suspicion is that the bacteria makes my mycelium weak enough to allow competitor moulds to take over fully colonised bulk substrates.
