Slant Test tube Suggestions?

I've yet to make my own slant and I think that should be my next goal. What do you guys/gals suggest for test tubes and do you suggest buying a 45 degree angle rack?

I've been using these test tubes and the work awesome and are very cheap

For a slant i just use a random sheet of metal propped up on one side by jar lids, I prefer my slant angle to be very low to maximize surface area of the agar.

I like mine, they are pp 50 mil centrifuge tubes. I use 25 ml of agar. I use no pour. Just fill them leave lid a little loose. Stand them in my filter quart grain jars, pressure cook,as soon as pressure is off. Take jars to flow hood , remove from the quarts and lay on upside-down styrofoam plate, make sure the lids are still loose or they will create vacuum and deform. When cool seal. These have huge surface area. Oh make sure you precook agar or it will separate.


My hope is these will last several years.

I like having the rack because I just fill the tubes with the agar while they are sitting in the rack and then place the rack into the PC, pull it out in front of the hood and angle the rack over, I ussually use the weight from the pc to angle the rack.

i like the 50ml centrifuge tubes that quadman posted. the opening is wide and easy to work in. if i remember correctly, the ones i purchased are autoclavable (including the lid). i don't have a rack, i just place the slants on a short stack of tongue depressors in front of the hood to cool.

Yep, that's why I posted my Tek fully autoclaveable . About 60 cents a piece. Enough agar to not dry out for years.

those ones i posted on amazon are also autoclavable, I prefill them before  loading in autoclave .  For more info on this method check out mephisto's master slant tek on mycotek. not sure if linking to other forums is considered faux pas here

The caps on them slide on and off so transfers are much easier. I clean and reuse them as well. had some for years.
As for slants, I fill them 1/3 for a wide slant then just put them all in your tube rack and lean them against the lip of your hood, a baking rack ,jar lids whatever you've got.

I think gr0wer uses ~1mm agar cubes suspended in distilled water for long term culture storage.

If you're using that then the tube size isn't so important ... in fact, it probably doesn't need to be a test tube at all ... just an airtight container.
