Yo I'm thinking about emailing David Nicholls at Purdue for a PhD deal
    #17728379 -

Ya know the guy, deeply fascinated by psychedelics bringing it up to a very high standard academic level.

So anyway I've given thought that since I'm overly obsessed with drugs I might ask him if he can accommodate me in his lab to study the second messenger systems that relay signals from the 5-HT[2A] receptor and how that contributes to the glow of the psychedelic experience.

I've got an outstanding academic record but I don't have a masters degree. In the UK its pretty normal to get into a PhD straight from your BSc and I'm wondering if thats how it is in the US or you need your masters?

Anyway its just a thought I have, nothing to commit to seriously at this point.

Good luck to you. It is not uncommon to get accepted into a PhD program with only a bachelor's, especially in the sciences. If you've got a plan and enthusiasm to carry it out, it's definitely possible.

This, but there's no need to e-mail Nicholls. That might even hurt your chances. You probably shouldn't mention psychedelics to anyone during the admission process.

Well I won't go all druggie-like on him

If I were to commit to this I would sit down and spend countless hours researching this particular topic and maybe even write my own project proposal. There was something about LSD and psilocybin both binding to the same receptor but inducing very different second messenger signals. Or different phenetylamines having unique second messenger profiles when the chemical signal is relayed from an identical receptor.

Basically the question is why two different drugs induce different responses through the same receptor.

But yeah I have to sit down and think deep and hard on this one.

I think you should definitely go for it. You sound interested and able. This would be a chance to get on some hugely important research that's on the cutting edge and if you're passionate about it this could be an incredible, life-changing experience.

If you do commit to this please share your experiences, discoveries and ideas with us here! I love neuroscience, molecular biology and research into psychedelics myself so I'd love to read all about your work.

The thing is I'm well versed in molecular biology and biochemistry but not very familiar with the study of neuroscience and practical organic chemistry (tho its something I'm willing to delve into). But yeah my main interest is molecular biology, what happens inside the cell when you dose it with psychedelic drugs. I can tag, mutagenize and clone the receptor gene and do westerns, IFs etc with my eyes closed but wouldn't know where to start from if I were to synthesize a drug.

steric factors

It occurred to me that different drugs bind in their own way where each molecule twists and turns the receptor so that specific cytoplasmic loop motifs and determinants appear. Their combined effects maybe dictates whether the effect is more towards the hydrolysis of phosphatidylinositol or arachidonic acid release. But how and why?? What factors push it towards one or the other end?

Different drug, same receptor, different response :mindexpanding: thats a very interesting topic in my opinion.

Oh man that takes me back to my freshers year. Lonely nights with the biochemistry books while everyone else is getting shitfaced in the pub :hehehe:

Then I found drugs again :Feelstrippyman::hehehe:

LSD is a pretty big molecule with a lot of interesting stuff going on in its structure. Tremendous room for electron resonance and play especially when near mega proteins littered with "hot pockets" with all sorts of charges.

Could be some cool stuff to research for sure. I hope our country gets its head out of its ass some day

The responses are not due to actions on the same receptor. It has more to do with the partial (%) activation of different receptors and sub types of those receptor types.

Sub-types cause different reactions, and activate different pathways, based on their activation (dosage), and specific activation of other sub-types (percent of each sub-type activated by a specific chemical).

The receptor sub-types mentioned here are just a few of the known types. There are also subtle other sub-type reactions that are harder to follow.

I have done University research with scheduled substances, but not schedule 1, those are in a different level of government-burrowing-into-your-ass. Schedule 3 was bad enough with an FBI background check, and we needed an MD/PhD with a schedule 3 licence to oversee the project. If they find any indication that you may use or abuse a substance, even legally, you could be barred from this research. Anything form weed possession to public intoxication. They even check your credit, finances, and personal references to determine if you could be coerced into misappropriating a controlled substance. And the team you will be working with does not want anyone with them that will cause any extra/unwanted attention from the government/media. As a student/PhD student, they are usually more lax. Some of the above may be more dependent on your employer.

The moral of the story - look squeaky clean even if you are dirty...or like to get dirty.

You should not need a masters coming from a decent University in the UK. I got accepted to Purdue with just a BS, as did a few of my friends. You seem qualified enough.

Two thoughts:
You seem to be at least a year into PhD work already? it may be in your best interest to finish your PhD and then do a post doc with Nichols. That would be up to you. You may also qualify for a masters soon, and then transfer. Generally, if not always, grad school credits do not transfer, and you would have to start over.
Get to know the man and the research, and possibly visit or correspond with him and/or members of his research group. That would greatly increase your chances of being accepted into the Purdue PhD program. Find out what it is like to work for the man.

If you want to work with the man to get a PhD, find out what departments he is capable of recruiting from. Each department has their own admission criteria. Find out what the PhD requirements are, and what protection PhD students have from hard-ass advisers that like to keep students as slaves /extended indentured servants.

I had a friend at Purdue that decided that shooting himself in the face would be less painful than trying to finish his PhD in chemistry. Poor guy. Before he went there, we would have debates about "is it better to be a big fish in a little pond, or a little fish in a big pond". My contention was that little fish are more likely to be eaten. Purdue is a pretty big pond, and he was eaten by bigger fish.

It's a pain in the fucking ass, especially if you're trying to make something novel.  I wish I didn't waste years of my life in an orgo lab and just stuck with biochem doing black box chemistry with fancy equipments   It's not all bad though, what makes it worthwhile is when you actually make what you're trying to make and in appreciable quantities.

As far as skipping over the MS, that's pretty much what everyone does anyway, it won't be a detriment when applying.  Isn't Nichols emeritus now?  He probably won't be around much (hardly ever) if he is so take that into account.  A good PI is a nice thing to have instead of a bunch of lousy postdocs.  Although having no PI present will significantly lessen your stress levels.

Same thing happened to one of my buddies, worked in the same lab as me too.
