Many thanks for the links, Mycelio !

I find it very encouraging that storage under sterile water has been used since 1985 for the largest collection of wood decay fungi in the world, maintained at the Forest Products Laboratory, Madison, Wisconsin, USA (Burdsall and Dorworth 1994).

Also, many thanks again to Terry M for kindly providing these references!

Last year I wrote to Jessie Glaeser, at the Center for Forest Mycology Research (known previously as the Forest Products Laboratory). She had relatively recently written something about this technique. I asked her some technical questions, and about the current status of the lab project. Unfortunately, I got no reply. Not sure if anybody is still maintaining the distilled water cultures.

The Bursdall and Dorworth technique was one of those I tried out. It was kind of fun using the blunt end of a sterilized glass pipette to stamp out disks. But it didn't work well for some species with sticky mycelium. You frequently got an agar plug stuck in the pipette, and if you continue with this pipette, newly cut plugs would add themselves to this plug. You've immediately got to switch to a fresh pipette, but then on the first or second try, another plug gets stuck in the pipette, causing you to have to abandon this one, etc. Meanwhile, you've wasted mycelium real estate and plate exposure time.

Another disadvantage to the sterile distilled water techniques I tried was anticipating the need for a finite number of samples. When you fish out a mycelium cube or plug, it's gone. I found that cutting up a large number of samples is tedious, and adds to the contamination risk by having a plate exposed to even sterile air for a long time. So if you use the samples regularly, a very long storage life doesn't really help much. You've got to reculture it anyway.

Regards,
Terry

I just wanted to add that shitake do not like to be stored long term in distilled water storage. Zero survived at just over a year. From experience. Oysters and Cordyceps are doing pretty good at 3 years now. I pulled a few about a month ago and they are doing well.

The samples should be very small 4-5 mm and you should put 4- 5  per tube. Don't give them lots to eat.
