Stronger mycellia after continued reflasking.

My Calocybe Indica myc used to be aerial and very slow. I wasn't aware back then of degenaration so it was reflasked many times. But now, they're as fast as my oysters and rhizomorphic. It fruits so it's not contam. Of course I'm ecstatic about it but I just don't understand. Any ideas?

One idea is that your culture used to have some contamination, and after successive transfers, you have removed them and purified the culture.

Another idea is that you had a mix of C. indica strains in your original culture. Now that you have done multiple transfers, you have selected out the strong ones that made it to the growth edge the fastest.

Other possible reasons are higher sugar levels, causing slower and more aerial growth back then, there may have been an initial malnutrition, temperature may be a few degrees higher now and so on.

Degeneration and senescence would only turn a vigorous mycelium into a slow and weak one, that doesn't fruit properly. These terms usually mean that somebody damaged mycelium by treating it badly and didn't understand it. Of course it is still important to keep the youngest cell lines for later use.

Terry, I'm pretty sure it wasn't contammed. The mixed strain is a possibility.
Carsten, the younger lines are still aerial and slow in the same media and temps.

I would go with the mixed strain idea and selection of the strongest strains

Then the initial culture should have shown uneven growth on agar, with faster and slower sectors.

Thanks mycelio. I read months ago somewhere that you have to choose rhizomorphic growth and that's what I do everytime. But I never thought it was a factor of strain difference. Let me dig up some old flasks and I'll post it tomorrow.
