Petri Dish -10% success
    #15094632 -

hey there folks!
i do not know what to do anymore, it seems like my petri dishes gets ALWAYS contaminated. ihave around 10$ success and i dont know why.
heres the thing: ive build a new glovebox and before i pour the PDA into the dishes i sterrelize it for 30-40 min + bath the petri dishes in bleach for 2h.. then i put everything into the glove box seal it and spray shit load of bleach again.
than i w8 15 min for it to kill all the bacteria pour the PDA into dishes get them inside a smaller plastic box and seal it as well.. only then i open the glove box and transfer the small plastic box with the dishes into my fridge incubator which been sterelized with bleach earlier... still after 1 week it gets contamed without me even getting to the stage of open it ONCE or even open the fridges door...
needless to say i close all windows and have no air float while im doing all of the process.

tbh only time i succeded was the 1st time i ever tried lol since then everything goes downhill.. no idea why.

any ideas guys?

you have your info wrong, bleach does not sterilize

your petri dishes need to be sterile and they come in a bag ready to use when new, no need for bleach

try an get a sauce bottle to pour your agar in to the dishes
work fast but try not to make to much turbulence with your arms inside the glove box

i use a still air box to do agar work since i have big hands and glove boxes wont let me move my hand freely

are you using parafilm on your dishes ?

bleach doesnt sterelize? i know it killed bacteria, isnt that good enought?
and the dishes coming in sleeves indeed but there like 30+ dishes in 1 sleeve and i use like 4-5 at a time.. does it mean the other get contamed right away?
- whats parafilm? sry for the nub aquestion:D

bacteria aren't your only problem. Mold too.

Parafilm is a stretchy cohesive film that filters air for GE in Petrie dishes and test tube slants

so should i be using parafilm instead of the petri dish lead than?

remove the 5 petris you are going to use from the bag they came in. seal up that bag until you are ready to use the again..... it is more than likely you are getting contams from when you put your hand into the glove box or while you are cleaning your dishes.

if i where you .... i would grow a nice batch of mu.... sell them to my friends and then use that money to build a flow hood.

try using a wine bottle to poar dishes.

I'm surprised you're getting 10% success.  I would expect zero.  You're doing every single step of the procedure wrong.  Watch the petri dish video clip to get an idea of what you should be doing.  It's only a sample clip taken from the full release on DVD, but should give you the idea.

let me be more specific: i used to do everything like u show in that video except i dont have a flow hood.
after each and every petri dish got contamed i thought the reason might be that my sterile sleeve of petri dishes got contamed becuz i used some of the dishes and stored the rest for later, thats where i decided to let them swim in some bleach for couple hours since i guess  some amout of air and bacteria reached them after the storage process.  then ofc i put the can that holds the dishes inside (with the bleach) and a bottle of bleach spray+the sterelized agar jar inside my glove box, sprayed shit loads of bleach, did the pouring .stored in another sterile plastic box and got it out of my glove box and inside the incubator.  so how the hell it got contamed>.<

I'm sure they were cleaner before you opened them up and put the bleach on.  Next time, make half a liter of agar and fill the whole sleeve of 20 dishes.  Then, put them back in the sleeve or place three to four dishes per ziplock bag, and put the extras in the refrigerator until needed.

hmm.. solets say i wanna try and build a flow hood. and blower is too expensive.. can i use a regular air fan?

another thing i cant understand how could the petri dish be cleaner before i put them inside the bleach . doesnt bleach kill everything?

No, bleach will not kill everything. It is used to disinfect, not to sterilize. Sterilizing means removing literally all microbial life. Disinfecting something is less effective than sterilizing, and leaves things like bacterial spores alive. Again, bleach disinfects, it doesn't sterilize.

They radiation sterilize those petri dishes, they cannot be cleaner. After you use bleach you rinse it out don't you? You water isn't going to be sterile

You can use a blower/motor combo taken from a residential furnace system
as I did for building my flow hood:

Mine cost me about $100 plus some scraps of plywood to build.  I picked up a medical grade air filter (12" dia by 12"high and 3"thick) at auction for five bucks (old lady used it for a week and could not stand the sound running at night in her bedroom).  It has a 99.99% .02 micron medical grade HEPA filter and a good blower in it.  I built a box on top between the filter and blower and and got two new 99.97% .02 micron HEPA filters (2"thick) for air filters at Home Depot for $35 each.  I joined them with silicon and placed in front of the working area (scrap of Corian counter top).  It gives a work area of 19 1/2 inches wide and 13 inches high.  The medical grade filter is used as a pre-filter and in under the box.  No contamination in a couple hundred petri dishes and many jars and bags of grain.  Not pretty but it works and is more than sufficient for me.

so lets say i dont have blower and cant find any. will it be fine to use 12X12 cm normal 220v computer fans ?
and shud they be facing down from the cilling or facing front from the back side?
+ its seems hard to calculate the fmc of couple of them together any1 have an idea?

No that will not work. Call around HVAC and heating and cooling places and
they more then likely will give you or sell you a proper blower and motor
from a furnace.
