LC jar concept...

I've been researching some LC jar teks but haven't found exactly what I'm looking for, so I thought I'd throw this concept out there. It's a pretty simple 5 step process to convert 1cc of spore solution into 100ml of liquid culture.

Make LC vessel
  • Take a half-pint canning jar and drill two quarter-inch holes in the lid
  • With those holes, create two self-healing injection ports using RTV or silicone (left to cure for 24 hours)
  • Sanitize jar and add 100ml of preferred LC solution, a marble or glass shard(s), and affix lid

Create vent syringe
  • Remove the plunger from an empty syringe
  • Fill barrel with poly-fill
  • Replace plunger enough to seal the end of the barrel
  • Wrap syringe in tin foil

PC both items for fifteen minutes @ 15 psi (allow to cool).

Inoculate Culture
  • Place the sterile LC jar ,wrapped poly-fill syringe, a sterile spore syringe, and an alcohol-soaked swab in a still-air box
  • Swab both injection ports with alcohol
  • Unwrap poly-fill syringe, insert needle through one injection port and remove plunger
  • Insert spore syringe into the other port and inject 1-2cc's of solution
  • Remove both needles and incubate sealed jars

Create LC syringe
  • Place sanitized LC jar, sanitized (or sterile) poly-fill syringe, and sterile empty syringe into still-air box
  • Vent jar with poly-fill syringe as was done during inoculation
  • Insert sterile empty syringe into other injection port to aspirate inoculant and fill syringe


The concept behind it is pretty straight-forward: it provides clean gas exchange and vacuum release to the culture, yet it also allows one to agitate the culture freely during colonization. With other LC jars that employ poly-fill'd tubes, one must clamp the GE tube shut while agitating to prevent soaking the filter material. This new method removes that variable keeping the jars fully sealed while agitating, but still allows filtered air to be drawn into the jar when aspirating the culture into the syringe. One can also provide gas exchange to the culture at any time during colonization with the vent syringe (provided the needle is flame sterilized first).

As far as I can tell, this method will create a highly viable, fully sealed LC vessel with decreased risk of contamination. If there is anything that I have overlooked or if I am off my mark in any way, please chime in! And if this concept has been posted/proven/disproven already, please accept my apologies.

It's been done, it will work. :thumbup: I think it works better with a special lid, but that's just me.

Also- props for a proper outline. Lots of folks just post a list of tasks, you seem to understand it's about goals instead.

I have not made lots of LC's but I tried both jar styles, with and without polyfill filter, and I prefer without.

It allows to shake your LC without worrying about the filter getting wet.

That's the benefit of this LC jar, you can shake it like crazy and never have to worry about getting any filters wet because it sports two fully sealed injection ports and no open tubing.

Yea, I like those lids. I guess our concept relies on materials which are non-specialty items ie: all materials are available at any big box store. I think this concept came about because those are the materials that happened to be laying around the house at the time. I've also considered the benefit of plastic lids as well, as they have the ability to be microwaved as well as PC'd.

I guess all that is left is to try this method and see how it compares to others in terms of ease of construction, material availability, etc. Will report back!

id just make a magnetic stirrer out of a hard drive magnet and a pc fan. there is a tek on this site. it works marvelously. also alot of data suggests useing smaller jars for lc. you can make a lil.. let it grow.. and use it up. this way you use it all long b4 the lc has a chance to get bad while it sits.

Yes, a magnetic stirrer added to it would be a magnificent bonus. Maybe that will be incorporated in the future.

this is what i built and it works fairly well. TBH tho it only works if its a babyfood jar.. still a baby jar isnt a bad amnt of lc if you need more just make several jars then if one contams...meh (i think labeling would be huge) I plan on building a new one in the near future with a small regular fan and maybe a stronger magnet if i can find one maybe then it will do the bigger jars.

hehe... i can't help but feel called out......:grin:
although I still stand by my checklist method of doing things...... all the info is important, but if i dont have a checklist-style paper, I forget a little thing here or there, and it messes me up
