isolating a single genome

ive been thinking about doing this for awile but im a little confuses. so basically to strat it all use live tissue? stem work better than cap? and vice versa

Good question! Furthermore, I'd like to know what would be the best route to go if one would like to clone a mushroom using live tissue?

A typical monotub grow of a multi-spore rye spawn with some Golden Teachers: inoculated on day one and saw first signs of growth on day 5. On day 17 jars were 15% - 30% colonized so shook em up. On day 25 I mixed the spawn and sub in the monotub and on day 50 harvested the first flush. Just under 4 ounces dry in 1 month 19 days!

You have to use a sample that hasn't started dropping spores yet, obviously, or you can never be sure that your biopsy isn't contaminated with the fruit's own spores. Pick one specimen you deem fit for cloning before it breaks the partial veil. Pins work very well for cloning, too. Use the whole pin. If you are cloning a more matured fruit, take a biopsy from the stem.

After I pull the pin should I drop it in some rye seed or what? Maybe isolate it on some agar? I think I remember RR saying something about even live tissue contains other cultures.

Best to work on agar. mushroom fruitbodys grown from multispore can be made up from more than 1 set of ginetics. Its quite possible to have a tissue sample still produce sectoring.

While it is possible to drop a tissue sample into substrate and it works, theres still the risk of contamination. With agar contamination doesnt mean failure as you just transfer away from the contam, plus you can further isolate the tissue sample.

Using a pin for a clone is a good choice, at the pin stage mycelium is still growing rapidly and will colinise a petri dish pretty quick, often outruning any contaminations that may be on the pin.

You can also start from spores on agar and isolate until you get single sector strains and then test each 1 to see how they perform. Starting from spores takes much longer and involves alot more transfers to get to a single sector isolate and then you still dont know if it will fruit until tested.

You can then start getting more complicated and isolated monokorytic mycelium (mycelium form just 1 spore that hasnt mated) and use 2 different ones on the same dish and have them mate on the petri dish to create dykyrotic myc thats capable of fruiting. Very interesting just watching it happen on petris, seing the 2 grwoths of myc meet then a third growth start from the middle :-)


You can cross 2 strains on agar too by transfering 2 different strains onto 1 petri dish and see if they combine.

so if i use a pin then the resulting myc growth would not be a single genome. by cutting out sectors and transferring them to another petri dish i could isolate a single genome. how do i know what sectors to cut out and transfer though? would it just be the best looking part? like rhyzomorphic growth? anyone know of an in depth text for this please post
