hi folks! my wookie friend has a question... he wants to know if its possible to use eritromicine in agar mediums and if yes he wants to know if its ok to use with spores, unlike h2o2... thanks everybody.

As far as I know erythromicin does not survive autoclaving, which is why the antibiotic of choice is gentamicin sulfate (which does).

And yes, spores will germinate with antibiotics, which only kill bacteria.

What sort of chemical stimulates germination of spores? I had a fresh spore print of Agaricus Augustus and I innoculated several petri dishes with spores. It's been almost three months and still no germination (nor contamination). Spores of other species on the same exact medium germinated just fine.

I know it can be done but I haven't been able to get clones or spores of Agaricus augustus to do anything at all. Try adjusting the pH.

i think you might need activated charcoal in order to germinate these spores

I've encountered similar discrepancies with germination between species. I have been trying to germinate spores on plates containing a mix of ampicillin and kanamycin - and puffball spores germinated fine, while oysters and malabars didn't germinate at all. Weird

well i cannot tell you if this is possible but i can quote something
anno once mentioned on the topic of antiobiotics.

he said not to use them unless you have to,
like saving a supa precious culture you only got one contammed plate
or something alike.

otherwise one shall never use antibiotics,
since if u happen to grow a contam, it will be resistant and your
going to be in some serious trouble.

That is pretty much exactly the point.

Imagine growing out a pathogenic species that is also resistant. If you do not destroy the culture, you unleash it on the world (although other antibiotics may work, it doesn't matter - you have already made it resistant to one). I do not think we need to accelerate evolution of bacteria in this way. It is bad enough with all the antibacterial soaps, kitchen cleaners, and other things we use. These products have negative impacts by causing bacteria to become resistant to them, and by lowering our exposure the bacteria (thus limiting immune system stimuli). USE BLEACH TO CLEAN! Hot soapy water to wash with...


Use antibiotics only when you need them (contaminated spores would be the primary use, other isolation techniques usually work with live mycelium cultures). And if you use antibiotics and still grow a bacteria culture: Destroy the culture by autoclaving it, do not simply throw it away!

Antibiotics are good for things like slants and long-term culture projects (i.e. strain isolation). They will not hurt anything but there is no reason to use them for short-term projects.
