Man yeah. Same here. Lacking motivation. Barely culting. Old tubs and jars stacking up. Spending time and money on non-cult related activities. šŸ˜†

I have grain jars and AIO bags I prepped several weeks ago sitting here unused. They look fine, I'll use them.

I still need to clean the lab, sterilize petri dishes and pour some freshies.
But I've dumped and washed everything that had gone fugly, so the worst tasks are done.

I'm pulling out of the slump.

I've been lagging, flagging and slumping lately. Low motivation. When I have the drive, I start something, then let it go to shit. Got through that, ran 2 batches of failed spawn, blaming new grain. Started new plates a week or so ago, replated the failed cultures, streaked some new spores (ochra/semps) from friends, all looking great. But now I'm leaving tomorrow for a week, I don't have time to do any transfers. Will likely need to start over when I get back. But I have other projects that will need to take priority, like heat for winter that will cause an early end to my season, again, as nothing likes to grow at 47⁰f.

Feels. Went out of town and the leading edges on my agar are about to reach the edge of the plate. Feeling under the gun but also tired and got other shit to do.

Curious what "start over" means to you in this context.

Y'all trash a plate if it reaches the edges of the dish? Is that a matter of being exceptionally particular about things or is there some risk involved with letting it grow out that far that I'm not aware of? I know I've seen it recommended to do something with the plate before it overgrows the plate, but I didn't get the impression it was *that* much of an issue.

Not an actual issue for me and I'd never trash a healthy plate. I've just been trying to grab that good looking leading edge while it still stands out visually from the rest of the plate, and it's also motivation to keep things moving along so I'm not dilly-dallying.

I imagine getting to a plate early matters more with MSS than a transfer.

If a transfer plate looks clean a week in, there's no reason to think it's dirty a month later.

With MSS, there's a good chance that not all the mycelium you see is actual mushroom mycelium. Letting it all grow together makes subsequent transfers much more difficult.

If I'm wrong, someone please tell me

I was talking about the new germination plates. They all germinated great, but they aren't clean. So when I get back in town, they'll be worse and will need a do-over.

But I just had a look, 1 of 4 of each variety appear clean, so maybe not a complete mess!

I gotcha. Man...this is like the 3rd time today I've replied to something I misunderstood. I think my friggin brain is just misfiring today or something. Yes, I could see how letting the contaminates march along for an extended period of time would be problematic.

I mean, I gotta be real, at the end of the day we're mostly making up stories that correspond with what we see as giants looking down on a whole world from very high up. So while I'm inclined to agree with what you said there, I also have to acknowledge that I don't necessarily have any idea what I'm talking about.

But yeah, at this moment I have a bunch of cloned cultures- many are clones of coones- and I'm just letting them sit on a shelf and dry out until fall. I've had pretty good success with ripping dried chunks of agar out and getting clean myc growth from them. Doesn't always work, of course, and a lot depends on how long they've been sutting around and whether I'm able to get in there with a scalpel or pair of tweezers and pull off the transfer without any screw-ups. But I definitely wouldn't expect to get away with any of this with germ plates.

I've never had as much success with squirting spore solution onto agar, streaking from print has tended to work out better for me. I haven't really done enough of either to know what I'm talking about, but my story there is that having that solution all over the plate makes spotting germination and separating it from contaminants a lot harder.

My personal preference is also to avoid excess water on the plate for that reason. Not to mention water sloshing around on the plate is about as good of a bacteria vehicle as you're gonna get. Everything looks weird to me on a wet plate. šŸ¤·ā€ā™‚ļø

I guess I was incorrect with my terminology. By MSS, I meant 'from spore'. I forgot that the last 's' stands for syringe.

For me, I try to be quick with transfers when working with any type of spore and I just want to check a transfer plate for clean growth within a week. If the transfer plate looks good, then I might expand it to more plates, put it to grain, or just forget about it for a while. The urgency is no longer there.

Haha I totally read it as just "from spores," too.

Yeah, I guess I've just gotten stung so many times that I'm all about redundancy, and I'd rather take an extra month making sure cultures are clean than waste two growing them out and having them go "meh" on me.

For me, I guess with any new transfer plates- whether they're from spores or just old dried-out shit- I want to do a couple transfers where I'm catching fresh growth before the plate is colonized. I figure the first transfer still has the old material in the plate, the second transfer has material that was in the same room as the old material, and the third transfer didn't have any direct contact with the old or germ plate. Once I'm comfortable with the number of transfers I've done, then yeah, it kinda goes in the neglect pile for eventual dropping on grain, development and collection of further plins, and/or long-term storage.

Well, for me, it's like this:

When I'm trying to make sure a culture is clean, and/or narrow down and bifurcate genetics via selective transfer, then ai consider that effort a failure if I wait too long and no longer have the ability to cut from the leading edge. If I'm confident in a plate and I'm looking to drop some of it onto grain and/or let it colonize and pump out plins for cloning, and/or just expand it out into a bunch of extra plates, then I don't worry about that.

If a plate grows out and I miss the window to take the leading-edge transfers I wanted, then I just consider it kind of like going back to square one. I don't toss it, but I do re-start the imaginary transfer clock, and assume that my myc is back on the starting line right next to all its microbiological frenemies.

But yeah, if I'm in "rip and run" mode, I ideally want to catch the new myc as soon as it's practical to get in there with my blade.

Yeah.

Well, I don’t know, he was one of the first "celebrity" musicians I knew about as a kid. First song I learned to play on guitar was Iron Man. Grew up with all the "Satanic Panic" shit, and gradually took in the entirety of his discography over the course of my adult life. And he had a whole second career as a sort of reality TV star/public figure. It was a little corny, but it kept him peripherally "in" my life all the while, with his voice still always somewhere on my playlist.

And unlike most celebrities/musicians/public figures, he always struck me as a genuinely real and nice dude who didn't get into any of the real sordid shit that most people in his position did. Ok, well, not always: as a kid, I believed he was possessed by the devil, and bit a real bat's head off on purpose, and glorified drug use, etc. But, it's like I spent my whole life learning more and realizing that was all just an overblown part of his public persona, then entered this phase where I'd be arguing with my wife about this sort of shit or whatever, and then, wham, he's gone. And even if Geezer wrote most of Sabbath's lyrics, it's still Ozzy's voice in my head. And oh, how they've aged like wine.

Anyway, I guess, like Lemmy, I FEEL like I knew him.

Either way, it's the end of an era, and a reminder that none of us are getting any younger.
