hi there!

so im starting this thread now before Ive even started the experiment, its going to take a long time to get any results and even longer to form a conclusion. any and all help would be greatly appreciated. i have a degree in biology and minor in chem but Ive only taken one botany class, nothing on micropropigation and im new with hormones :blush: for plants that is.

Can 6-benzylamino purine be used to create shoots (which do not usually form) from Salvia div. leaf only cuttings?

Today i will take a large leaf from one of my salvia plants. i will cut a cm from the stem of the leaf and then cut the leaf into sections that are 1-2cm wide and 2-4cm in length. Small cuts will be made on any portion of leaf vein that may be present. Ive noticed that the vein in the leaf is usually were the roots originate from. the cuttings will be wetted with tap water and then 2/3 length wise the leafs cuttings will be covered in powder rooting hormone.

I will then place 1/2 of the leafs 2/3 of the the way into small plastic cups with dirt and a second cup on top for humidity. the others will pe placed in individual cups filled with tap water and a second cup on top for humidity. they will all be labeled and dated.

I have rooted multiple salvia leaf only cuttings successfully in dirt before. i have not rooted any salvia using only water at first. in my experience (probably about 10 leaf cuttings to date) the leafs form roots in about 2 weeks and grow very slowly. the longest i ever kept one after rooting was probably 4 months and it never sent off any shoots.

Ok i ordered a gram of BAP pure powder today. it should arrive soon. as soon as i have confirmed that the leafs have roots  i will apply the BAP to the leaf surface near the air dirt interface. im thinking of using between 1 and 3% bap mixed with lanolin. i dont think scratching or otherwise harming the leafs would be of any benefit. if i can apply the BAP with out the use of lanolin i would prefer it. a measured amount of bap solution pipeted onto the air dirt interface of the leaf may work and Ive also heard about people injecting which may work where a root and the leaf meet. anyone with out with out experience using this hormone please chime in.

I will keep the humidity domes on the entire time. after seeing the amazing things that people have done with cacti and BAP i really think that there is some hope of this working.

If it were to work, we could take leafs and turn them into at least 4 cuttings though several times this may be just as realistic. and microprogiation methods involving sterile technique as well as agar and other hormones would not be necessary! Ive successfully rooted leafs from salvia plants no larger than a square centimeter.

i have also rooted (all of my salvia clones root in dirt) a leaf that had a small section of a node included with it when removed from the plant. these usually do create stems. but that causes damage tot he plant and only results in one cutting per leaf.

so im starting this tonight. ill post pictures. i have to do everything from my phone so im sorry if things are a little messy, i can clean them up when i go into the library.

I would be very grateful for any feedback. salvia is very easy to root and currently it it a plant that i own, plus it doesn't create full plants from rooted leafs unlike p. verdis and others. however anyone with experience turning any leaf into a whole plant please chime in.

i understand this may seem sort of far fetched but please dont shoot it down unless you have some intelligent and backed reason to do so. i have not found any scientific articles that discuss this though there probably are plenty of them out there.

ok i admit i didnt search very hard or long for articles that had to do with this procedure. below are addresses of several articles that Ive found that have similarities with the method im going to try out.

the midrib of the leaf is the region that i think the shoot will most likely come from. therefore all cuttings must contains some of it.



the best way to include it and produce the most cuttings from one leaf would be to cut the leaf perpendicular to the mid rib. everyone agree? that way part of the lamina is below and the other portion above the dirt while the mid rib is at the air dirt interface where it can produce shoots and roots and easily have the bap applied to it.

i completely forgot to mention that i will have these cuttings under lights. they will be in my closet with the mother plant the leafs came from. there are several 26w cfls and a humidifier. they are on a 16to18 hr/8to6 hour on off light regime

all the articles i am finding involve sterile media. however since salvia d leafs can factually root in dirt i see no reason that they can not strike shoots by simply applying bap to the mid rib

Hey sounds like a good test.  Looks like you'll have plenty of BAP to play with.  I have witnessed a Salvia d. leaf cutting fill up a ~3/4 gallon clear pot with roots and never send up a shoot and many other people have encountered the same results.  Keep us updated.

hey thanks for the interest! i will be home in an hour or two and il dry to convince my photographer girl friend to help take pretty pictures (ill try at least). i once sent Daniel Siebert a email about doing something like this and he responded saying he hadn't seen it tried though i dont remember exactly what else he said. ill try to round it up.

with the leaf cuttings you've witnessed, were they whole leafs or portions of one? did you notice where all the roots originated from on the leaf?

Alright phase one is done.

Last night I made a clone, I put the leafs in a plastic bag in the fridge. They seem just fine. Anyone have experience here? Couldn't get help taking photos so I have a bunch of grainy ones from my phone.


And now I can't figure out how to post pics from my phone... So I cooked some potting soil at 350 in the oven for about 20 minutes. I took cuttings from the leafs perpindicular to the mid rib of the leaf. I then soaked the cuttings in water with rooting hormone in it to make sure they were hydrated ok. Then I put root ing hormone powder on the leafs from the mid rib out to the edge of the leaf but only on one side of the mid rib.

I placed the now cool soil into the "pots." I used a popsicle stick to make a hole, inserted the half of the leaf with rooting hormone and then filled in the hole with more soil. Placed humidity domes on them after spraying it down with water and some neem oil to help fight fungi applied to the top layer of soil.

Pretty basic stuff. I also put some cuttings into a container with rooting hormone disolved in water. I don't have a bubbler at the moment so I will change out the water as often as I see fit. I'm hoping to use them as a way to know when its safe to check the in soil cuttings for root growth.

When the bap arrives (just ordered today and I'm gone this weeekend) ill update the thread with what I decide to do with it. If anyone has any recomendations let me know. I have pure 100% aloe vera gel, any idea if its ok to use with bap and on plants?
