Autoclave to replace pastuerization
    #899993 - 09/22/02 08:52 PM (14 years, 2 months ago)

a friend in holland's cat reports:

i have been trying to pastuerise dung/straw using the normal methods but i have found this too messy/problematic. i have access to an autoclave and i was wondering if i could simply autoclave the straw/dung at pastuerizing temperatures, 145 F?. would this work? i have autoclaved straw before at 121 F but have had some contamination problems. thanks in advance for your responses...

I am not real familiar with autoclaves but i suggest u make ur pasteuization temp 160-170 and that will help u to get less contams. u might also want to think of a way to keep ur moisture content at the normal levels. other than that i believe it would work fine.

thanks cubies4me:

i was going to check the pastuerisation temp before i did it...i'm sort of new to this..

i would soak the straw for 24 hours and drain/squeeze it before placing it in turkey/aluminum pans, covering in foil and placing in the autoclave.  i would think this would be similar to steam pastuerization.

thanks for your comments.

If you do decide to use your friend's autoclave.. Make sure you put your straw or any substrate materials in an autoclavable bag, open at the top to vent. The materials can break down into small pieces and when you go to drain the chamber.. the particles can clog up the drain.. causing mucho havoc and repair$.
I suggest when the cycle is complete, let pressure come down naturally. Ck and make sure there is no debris in the water. If there is .
then empty out the cooled water from the chamber by pouring it out manually into a sink instead of risking it flowing back into the outer chamber... I had a bag of casing explode all over the place once, it was a mess, and cost me 150 bucks for repairs.
Good luck..

thanks patzee:

the cat has autoclaved straw once before (at 121 C) with the only ill effect being that the lab smelled like cereal. he placed the wet straw in a turkey pan, placed aluminum foil over the top and taped it on snuggly. then he cut a hold in the foil and affixed a filter patch for air exchange/ pressure release. this is one damn smart kitty. the cat has tried using poly bags before (he has about 1000 of them) but the bags melted, probably because they were in a metal pan. i will recommend that he try poly bags in a plastic container.

i will tell the cat to be more careful though, as i've heard these machines are rather expensive and he would probably be expelled for destroying one.

the cat tried his experiment....

unfortunately, the autoclave only goes down to 100 C so i couldn't get the temps low enough for pastuerisation. the cat went ahead anyway and autoclaved at 100C, probably a mistake. he spawned on container of poo/straw with F+ and the other with some unkown strain..maybe Tazmanian... although i doubt it since all his other jars from the Taz syringe have shown no growth. (thanks LG!)

will update on progress...

I just bought one last week, a college was closing down a campus...and I work for the moving company that is moving them...verything is for sale now in the campus...and the Janator sold it to me for 5 bucks...I knew they were expensive, but I had no idea. This particular model is a few years old...but damn the new one is over 1000 USD....I am still amazed. The thing works too. I will try it out with some jars and make a growlog....cheers

i have autoclaved straw before at 121 F

Are you sure about this? Usually 121 is in C not F, which is the temp at 15 psi. I usually pastuerise at 160F and sterilize at 121C.

Thanks to Anno,
Autoclave-Basically a big pressure cooker, sometimes operating at higher pressure than 15 PSI, thus achieving sterilization temperatures above 250?F.
