Cloning Trametes versicolor ("Turkey Tail")

I have a few old firewood rounds which are sprouting turkey tail this year. I am very interested in cloning this. But the fruit bodies (as you know) are very flat which makes it difficult to get inside to the clean tissue.

Could this be a good place to apply H2O2 techniques? I'm thinking of picking an entire fruit body, rinsing it well under running water, ripping it a little bit (not completely in half) to expose the inner tissue, then a dunk in diluted H2O2 and then straight to liquid culture. I have no way to do agar at the moment.

Could it work? Or, failing this, I suppose I could attempt to grow from spores. Is T. versicolor a good sporulating species?

Baffled.

If you don't have the means to use agar, don't attempt cloning Trametes. 999 out of a 1000 isolation attempts will result in contamination. You'll need agar not only to see these contaminates, but to transfer away from these contaminates.

Haven't I read posts about success using H2O2 dunking of tissue and then straight to LC? Maybe that was with cleaner specimens? Or is it hit and miss in general? I'm not attempting to create a stock culture directly from this tissue, just multiply it a bit.

Okay. I was hoping there might be a clever trick to get inside such thin fruit bodies.

But how about this? Collect lots of fruit bodies (I've got tons) and rinse them extremely well. Then blend them with distilled water to make a thick slurry, and use this at a high ratio (1:1 maybe?) to inoculate some kind of substrate. Keep this in a container and incubate it outside under the same conditions it was naturally growing. There's still plenty of winter left.

Maybe I could colonize the substrate and then re-fruit, but indoors this time, producing much cleaner fruit bodies.

Yeah yeah, I should just make some plates...

Wild specimens (especially polypores) have a wide variety of fungi and bacteria growing among them. On or in them really. So these competitors will show up as well in sterile mediums like liquid cultures.

Why don't you lightly sanatize a piece of cardboard in some boiling water, allow it to cool and drain a bit and then sandwich a small chunk of Trametes in it. Then put that in a filtered jar or even a Ziploc baggie for colonization. Then you can expand that onto dowels or whatever else to be used for inoculating stumps, exc.

Hello vdlsar,
welcome to the Gourmet section. I've never cloned T.versicolor, but you need just a really small tissue to clone it.

You MUST obtain a pure culture, stock or not stock, to multiply it, or you'll multiply simply contaminants.

Cleaning something is LC is quite a miracle. You should clean the fruitbody with alcohol and then with iodine(perocide is too much aggressive, then take an inner tissue and clean it making multiple transfers (you have to take just the uncontaminated mycelium). You should have the antibiotic to add it into the agar (like gentamicin), to rpevent or reduce bacteria contamination. If you need it i can send you 0,5 grams for free that's enough to make 3+ liters of media.

If you don't want to use the agar, blend the T.versicolor with water and pour the slurry outdoor over a bed of woodchips and woody plugs in a shaded place. Cover everything with the inner part (corrugated) of a moist cardboard, and remember to water it when not moist. You could also put everything into a cardboard and watering it when doesn't rain from a reasonable time.

You could use this natural spawn to inoculate logs and similar..

If you want to produce T.versicolor indoor, go for the agar way, jars etc etc, if you want to grow outdoor use the cardboard method. In my opinion you should clone it on agar and isolate the pure mycelium, then create slants and store them in fridge to reuse when you want. At this point i'd create grain spawn to inoculate a bag of sterilized(better)/pasteurized plugs and with this plugs i'll inoculate some logs.

This will probably work with turkey tails, I wouldn't even rinse them and I would use just tap water. Spread you slurry on some fresh cut wood and put the wood in a shady wet spot and I think you'll have a good chance of getting some turkey tails. Might work with chips too, I may give this a try.

I'd think that would work as well. Turkey Tails are very hearty mushrooms and seem to spread easily.

Take the stem butts at the log surface and place on soaked cardboard or wood chips. Just one more fun way to skin a cat.
