Ok, I have read a lot of good things about liquid cultures with Karo. I found some teks here and there and read some posts about how to do it, but I must have missed this part:
What is the ideal lenght of time to let the mycelium grow in the water/karo solution before it can be mixed/used?
I just started 8 cultures today (4 ecuadorian and 4 creeper) to see how things go.

Alright, I just wasn't sure, because I had never seen anything posted about how long it is supposed to sit after mycelium growth. Thx

Wait until you have some heavy Myc growth in the culture. Hope you have a glove box, hepa flowhood, or clean enviro to work in? Liquid culture is a contam magnet.

I'm building a flowhood soon and my friend has a cleaner environment than I do, so I might just work out of his place. But the supplies for the flowhood should be here within a week or so.

When I see growth about the size of a quarter-or a golf ball, I then shake the hell out of my jar(with little pieces of broken glass in the jar to aid in breaking up the mycelium)and out of 100 ml of solution-I get 10, 10ml mycelium syringes. What I don't use, I store in the door of my fridge to slow growth for later use. This is a quart jar with 700ml of liquid,which makes about "70", 10ml syringes of liquid innoculant.More than enough. I also use this method to check the viability of my prints/syringes. I use 1 ml of a spore syringe to inoculate a karo jar,which leaves me with 9 ml left-if the jar doesn't contam -the syringe and print are clean. Better than wasting all the jars if I inoculated them right away and they ended up contaming. IMO,using Karo is the only way to go! My two pennies.

Doc, do you have a link to the _exact_ karo tek you use? I've tried karo a few times and it never seems to "take off." I use a tbsp. of karo for a pint of water, split that water into two pint jars (so they're half full or so), cover with tyvek, bands, pc for 30 minutes, let cool, inoculate, and wait. Latley, if I've been swirling (once a day) these slow starters I have lying around, but I'm not sure if this helps or hinders growth. I do have glass in the jars to break up the myc (as if I had any globs to break up ). The only thing I've noticed is the karo seem to colonize better at 80 degrees or even lower. WTF? Any insight you could share? Thanks very much!

My Karo looks like a couple of fluffy cotton balls on the bottom of the jar. Is that what I want, or is that contam?

I use 2 tablespoons of Karo for every 100ml of water. Pc for 15 minutes at 15 psi. I incubate my jars at room temp.Yeah, I just toss them in the cabinet and check back in a couple of days.I don't swirl my jars except for when I handle them. I prepare my karo jars just like my grain jars-tyvek+disc w/polyfill+band+foil. I store the ones that I don't use right away in the door of the fridge, that way they get swirled everytime I open the door and it slows growth to a crawl. They will continue to grow until all of the Karo/nutes is consumed by the mycelium. Hope that helps.

All of this has been a great help, thx all.
I prepared my solution with 1 tbsp Karo and 100ml of water in a glad 118ml tupperware container.
To sterilize I put them in the microwave with the lids half on and let cook for 1 minute, let sit for a few, cooked another minute, let sit again and cooked for 1 more minute.
After I inoculate them with 1cc of spores I'm just going to let them sit and look at them every so often to check the growth.
Does this sound pretty good, or does anyone suggest any adjustments?

One more quick question. Should I shake the karo mix before inoculation to ensure karo/water is mixed well?
