Don't we just call this cryovials ...?

I heard it called distilled water storage, water stasis, cryo vials, long term water storage, and water cultures.
I called it cryo vials but it's Castellani.

I consider cryovials to be a specific container. (Tiny, shitty ones imo)

To resolidify agar, i would pc at 15/18 psi for atleast 10m
Make sure you vent.
Ive never tested it, often wondered. I assume it'll be melted by the time it gets to pressure but as i said ive never tested it

Woah woah why are the tiny ones shitty!? I like to use the 2ml ones. You can get the pp5 cases that hold 100 of them and sterilize the entire case with them holding the centrifuges. And they're very easy to pull the piece back out if you cut the agar into a long rectangular piece.

Im not a fan of small stuff i wanna keep sterile. Easier to fumble or get in the way of for someone who rarely uses em.

Like a little dime bag ziplock? What im supposed to shove my thumb in that opening to peel it open? Great way to stay sterile.

Id rather receive a full plate in the mail than a smash wedge.

I want my cultures easy to access without concern, and an opening the size if my finger or smaller is a nightmare for me personally

For me personally, yeah I consider that too much moisture. I don't mind a tiny bit of condensation above the grain line or just barely below it due to temp swings but down in the grain like that is too much for me. Not saying you can't send it and get a flush or 2 but I had bad luck with things looking like that.

Thank you for your explanation, much appreciated! Sharing your expertise helps me a lot on my learning journey:hatsoff:

I guess you actually meant reliquify or desolidify

Empirically tested, 10 min venting + 10 min 15psi, worked as a charm. Gut feeling is that I could have get by with less.

You can transfer mold multiple times but it stays mold.

Gaining the ability to identify clean mycelium is a skill that is often lacking with newer users.

This is also not something that can just be explained with words, visual identifiers are key.

Your mycelium is very disorganized and does not show a clear leading edge.

Imagine it on a flat 2d plane, healthy mycelium spreads across the 2d plane in a uniform, ordered manner.

The mycelium you have posted is fluffy and disorganized with no visible leading edge.

Hi,

great I do save some dishes from privously batches and i forget too show 1 sample from the last transfer.. Mi thought it is the slowest i don't make a pic and show it and maybe it's cleaner then the one you say they not clean.

Here is the DC Mac Dish from the Oct 8th the slowest one..

Here is a Dish from Jack Frost i think i transfer a piece from it too my LC. Sry the lid looks litte dirty I hope you can see a part i can transfer again...

Here is another Dish from 13.09 also DC Mac stored in a fridge.

And last one also Jack Frost this is the one i swap a spore print on it it's coming from a fridge and it's cloudy so impossible too you too see something but i show it too

What would you do from here? I hope there is a good Mycelium somewhere too transfer... I have know the 7 not clean dishes so i can empty them and PC new Agar that's not a problem.

Thanks guys.

I can't be the only one who has twisted open a coke baggie one handed while driving, shoved a rolled bill in there, and took a half gram rip while keeping the other hand on the steering wheel at all times?

Never really been my thing. Did mushrooms all summer when i was like 15 and never looked back. Had a few lines in my early 20s when i worked at a bar, but addictions always ran in the family so ive avoided harder stuff

Hey Guy's

I do new Dishes but this time i don't use any food grade colors the KI gave me the tip using activated carbon that is black and perfect for spot any contam...

Quote
In mycology, using activated charcoal in agar offers four key advantages that go well beyond mere aesthetics:

Visibility of contamination: Against the deep-black background, you can spot contaminating bacteria (often yellowish or slimy) or unwanted molds (frequently green or gray) at a very early stage—things that are easily overlooked on clear agar.

Easier selection of mycelium: Snow-white fungal mycelium stands out perfectly, allowing you to use a scalpel to precisely excise the strongest, fastest-growing mycelial strands (rhizomorphic growth) for further cultivation.

Binding of toxins: Activated charcoal acts like a sponge, absorbing toxic metabolic byproducts—whether released by the mycelium itself or resulting from dying cells—which helps keep the growth medium "fresh" and healthy for longer.
Inhibition of bacteria: Bacteria generally do not "like" the structure or the absorptive properties of activated charcoal. Fungal mycelium, by contrast, is completely unaffected and grows right over it, providing a mild antibacterial effect on the plate.

So i gave it a try and PC them in Aluminum Foil for 25min

This is a result

Would be Cool when you say which part of Mycelium i can transfer will do that tomorrow morning.
