I don't think you should worry so much about senescence. I think most people 2ho get worked up about it or suggest that the limits are nigh are conflating senescence with random contamination sneaking in. I think the only thing that really matters is separating new growth from old growth, because old myc will certainly senesce and introduce all kinds of crap into the mix.

But you also have to distinguish between the theoretical limit of telomere expansion and everyday myc-getting-beat-up. People tend to think in terms of how many transfers they made, but the mycelium doesn't know that. It just knows when there's new material to colonize. If I catch new growth on a plate 5 times in a row and keep transferring it each time, the only functional difference between trans #1 and transfer #5 is that the stuff in transfer #5 is completely separated from the old, beat-up myc that would be in plate #1 if it were still around. That's it. From the point of view of the organism, it's just strands growing in straight lines.

I don't know anything about suspending agar wedges in water, but I've had shockingly good luck lately with reviving old dried-out plates that have just been sitting at room temperature. Some over a year old. A few will show immediate signs of contamination, and a few others will just never come back, but most will send out new clean growth once that dry agar touches a fresh new plate.

I've got culture lines that are almost 10 years old at this point. All of them were "preserved" via a perpetual cycle of storage and use. Sometimes cold storage, sometimes room-temperature neglect. And never worrying about keeping the earliest versions around. Just ripping and running.
