Chronic Petri Condensation

I can't seem to escape condensation. I'm now using an IR thermometer for my pours and mostly eliminated condensation from my fresh poured stack on standby. But now today I did a few transfers, streaked a few and my plates that were clear before I started they're now starting to condense. I do my agar work in front of a hood so I don't know if they cool in the airflow then they're exposed to warmer temperatures in my room that makes them condensate? I've tried the warm/hot cup on top of the dish but it doesn't make much difference. Some dishes get so bad it's literally a tsp. of water droplets on the inside of the lid. I don't know wtf it comes from  :psycrankey:

Could it have anything to do with how tight or loose I wrap with film?
Storing them in a less humid environment?
What the hell can I do?

switch to no pour. the ge hole fixes excess hydration by itself :hehehe:

I have so many no-pours under my belt I sold out the stores in my area. You're right condensation wasn't really an issue with those but I dislike working with the high wall and low visibility. There has to be a way to correct this with petris.

I’ve been struggling with this myself ,last weekend I did 20 agar petri dishes
I poured them in front of the Flowhood left them for half an hour so they became the same temperatures as  the air,then I did my work and hardly anything fogged up

I get what you're saying but I'm past that struggle. I poured plates 2 weeks ago in front of the FH and they all turned out good. I did transfers on those same plates today in the FH, then moved them across the room to the rack where I keep all my plates and they're starting to condensate already.

Condensation only occurs when there is a dramatic temperature change if there’s lots of water then you’re agar is not set yet

Not directly but I have a lamp that's pretty warm and I keep a few tubs on a rack below them. I'm thinking it has to have something to do with temp and humidity in that area. Maybe I'll try moving them closer to the floor.

If u keep a few tubs below it then move them that's probably what's causing it. Also look for where your hot air comes in make sure it's not blowing on them on under. Use the IR guy u got. Check temps in ur surrounding room vs where you keep ur plates.

I was having this issue but realized my heater is blowing right under where I store them. Moved em now no problem.

Also u can just let the sit in front of the hood for awhile. I'm a slacker so I usually wait an hour :bongload: before wrapping. Usually only have the very top petri slightly condensate. Cup of water fixes that :thumbup:

Condensation spreads contamination when it drips and pools on the plate. Funny enough turning the plates upside down is an answer to bad condensation, not an issue with it.


Temperature differentials are the cause. My work area got colder than I liked and mycelium growth slowed. I build a little incubator and realized I could NOT use it as my agar plates got flooded. Even slight temp differentials can cause it. The answer if to temp control the room they are in. NO you can not just store them some where warmer. They need to be kept at close to temp at which they set up at post pour. So work on keeping your lab temp from swinging. You may need to get an oil filled radiator heater for that room, and keep it say 72 all the time. I say have this heater a couple degrees above what you have your house set at. This way when the home heat kicks on it wont jump above what the lab heater keeps it, but then the lab heater will keep temp when the home furnace kicks off.

Last year I had to use heat lamps on the opposite side of the lab, this raised ambient temp enough that the condensation was tolerable.

Also keep in mind that the cultures on agar produce heat via growing, so stacking to many dishes in a stack can cause some issues as well.

Plates are stored correctly with the agar at the top of the container. They are upside down if the agar is at the bottom of the container, and condensation can pool on it. The only time my plates are upside-down is when they are being poured and PC'ed.

I don't think that is a "rule". Cause we never had to keep them upside down in biology class, nor do I now and I grow a metric fuck ton. If there is no condensation, then there is no need to flip them with the lid down.

Flipping lid down and agar up does have a possible negative side effect. IF you don't keep them in another container and the Para film cracks, then your lid will catch any spores that settle down. Gravity will assist spores getting into the dish. AND YES Para film can and does crack and pull back from the plates, it happens to a lot of us. This is why mine are wrapped and go into Ziploc bags.

TheMadHatter420 is correct. Petri dishes (the true dishes, not other containers that people use in leu) are designed to always stay with the lid on top and media on the bottom. The design uses gravity to keep environment sterile, so nothing settles to the bottom. During my 10+ years in the mol bio lab the only time I saw upside-down dishes (that is with the media on top) is in the biohazard trash bag.

That was a hard and fast rule in all the labs I've worked in. Of course, those labs had relatively clean air, so the biological load in them was low. Either way, it'll be difficult for any contaminants that settled onto the lid to make their way up to the agar.

That's the difference between containment and isolation procedures. In isolation, you are trying to keep bad things out. In containment, you are trying to keep bad things in.

So long as you don't flip it agar down to do transfers. If you do your them allowing anything that fell into the lid, the option to fall onto the agar. Maybe you take transfers upside down? I have never tried so I can't say it is not done and not a technique used by some.

I just use a flow hood and the only time the dishes leave the flow is in a Ziploc bag incase the Para film cracks on me. As for condensation, I think it is best to try to stabilize temps in the lab so condensation isn't a concern.

Yep, my agar always is "upside-down", with plate tilted enough so I can see the under-side enough for me to place samples.

Testing of LC and other cultures is done with the agar on the bottom side, 'cause obviously gravity.

I use glass petris, no-pour. Best visibility and reusable. No need for GE hole. After inoc, they go into zip baggies. Condensation clears in a few days.

If poured in front of a flow hood, let set for a couple hours prior to use and they dry out some condensation, and if your hood is built right, that air is clean. SAB's are a total different story for agar.
